Alzheimer's disease is a neurodegenerative disorder characterized by cognitive decline, synaptic dysfunction, and neurofibrillary damage. Alzheimer’s pathogenesis is often triggered by oxidative stress, which induces neuronal apoptosis and tau hyperphosphorylation, leading to the formation of neurofibrillary tangles (NFTs). Since current therapies remain symptomatic, exploring medicinal plants as potential preventive is essential. Petung bamboo (Dendrocalamus asper) is known contains bioactive compounds, such as flavonoids and proanthocyanidins, with potential antioxidant and anti-amyloidogenic properties. This study aims to analyze the antioxidant activity of D. asper leaf extracts and its fractions in vitro and to evaluate molecular interactions of its bioactive compounds with specific Alzheimer’s targets through molecular docking. The research procedures included extraction, fractionation, DPPH and TAC assays, LC-HRMS profiling, and in silico simulations of GSK-3? and Keap1 proteins. In bioactivity tests, the crude extract showed the strongest DPPH antioxidant activity (IC50= 69,9 ± 7,62 ppm), while the ethyl acetate fraction exhibited the highest TAC value (117,76 ± 19,96 ?mol/mL). Conversely, the n-hexane fraction showed the lowest antioxidant activity in both assays. LC-HRMS identified metabolites from the flavonoid, alkaloid, phenylpropanoid, polyketide, fatty acid, and amino acid groups. Molecular docking results identified three compounds, 5,7-dihydroxy-2-nonylchroman-4-one, 12-OPDA, and ethyl ferulate, as multi-targeted ligands potential predicted to interact stably with both target proteins. These findings are expected to provide a scientific basis for future in vivo studies to validate D. asper leaves as a novel therapeutic candidate agent for Alzheimer’s disease.
