Salmonella
Meningkatnya
kasus resistensi antibiotik mendorong perlunya eksplorasi alternatif
halo plaque. Uji kisaran inang menunjukkan bahwa faga memiliki kisaran inang
yang sempit, hanya mampu menginfeksi Yersinia pseudotuberculosis selain
inang utamanya.
biokontrol.
Salmonella is one of the major foodborne pathogenic bacteria that
threaten public health worldwide. The increasing incidence of antibiotic
resistance underscores the need to seek more effective control alternatives,
one of which is the use of bacteriophages. This study aimed to isolate
bacteriophages from the Kali Code River that are specific to Salmonella enterica and to determine
their characteristics through host range and physicochemical stability tests.
Water samples were collected from the Kali Code River and isolated using the double
agar overlay method; the samples were characterized based on host range testing
against five test bacterial species, as well as stability testing at various
temperature ranges (4°C, 30°C, 37°C, and 55°C) and under various pH conditions
(3.5; 5; 7.5; 9; and 11). The results showed that five plaques were
successfully isolated, exhibiting the morphological characteristics of halo
plaques. Host-range testing indicated that the phage has a narrow host range,
capable of infecting only Yersinia
pseudotuberculosis in addition to its primary host. Temperature stability
tests showed that the phage remained active in the range of 4°C to 37°C and was
inactivated at 55°C. pH stability tests showed that the phage is stable at pH
3.5 to 9 with optimal activity at pH 7.5, while at pH 11 there is a significant
decrease in activity. Thus, the bacteriophage from the Kali Code River has high
specificity against Salmonella enterica with good physicochemical stability,
making it a potential candidate for development as a biocontrol agent.
