Antibiotic resistance has become an escalating global issue and poses a serious threat to public health. This condition is primarily caused by the uncontrolled use of antibiotics in the treatment of microbial infections. Vancomycin-Resistant Enterococcus faecalis (VRE) is one of the bacteria with a high level of antibiotic resistance and is capable of causing nosocomial infections. One potential alternative agent that can be used to control these bacteria is bacteriophages, a group of viruses that specifically infect bacteria. This study aimed to isolate and characterize bacteriophages against Vancomycin-Resistant Enterococcus faecalis derived from soil surrounding the TPS 3R Purwo Berhati, Yogyakarta. Bacteriophage isolation was carried out using the enrichment method followed by the plaque assay method. Characterization was conducted through plaque morphology observation, host range testing, and stability tests under various pH and temperature conditions. The results showed that the bacteriophage was successfully isolated with a titer of 3.33 × 10^9 PFU/mL. The bacteriophage produced clear plaques with distinct edges and a diameter of approximately 0.8 mm. Based on the host range test, the bacteriophage also demonstrated the ability to infect another bacterium, namely Listeria monocytogenes ATCC 7644, and remained stable at pH 4, 7.5, 9, and 11 as well as at temperatures of 4, 30, 37, and 55 °C.
