Antimicrobial resistance (AMR) has become a serious global health concern, one of which is driven by the increasing incidence of infections caused by Methicillin-resistant Staphylococcus aureus (MRSA). The high level of MRSA resistance to various antibiotics has encouraged the development of more specific alternative control strategies, one of which is the utilization of phages as biocontrol agents against antibiotic-resistant pathogenic bacteria. This study aimed to isolate and characterize phages with lytic activity against MRSA from environmental water samples collected from Kali Code, Yogyakarta. This study was conducted using experimental laboratory methods, including phage isolation through direct isolation and enrichment isolation methods, purification, amplification, titration, and host range testing using the spot test method. Phage characterization was carried out based on plaque morphology, lytic activity, and infectivity against several Gram-positive and Gram-negative bacterial isolates. The results showed that phages were successfully isolated from Kali Code water samples after the enrichment process. The isolated phages produced small, clear, circular plaques (clear pinpoint plaques) on the MRSA bacterial lawn. The amplification process successfully increased the phage titer from 3.67 × 107 PFU/mL to 1.27 × 10¹² PFU/mL, which was categorized as a high titer lysate (HTL). Host range testing demonstrated that the phages were only able to lyse the original MRSA host and did not produce lysis zones on other tested bacteria, indicating a narrow host range. This study demonstrates that environmental water from Kali Code has the potential to serve as a source of lytic phages against MRSA and may be further developed as phage-based biocontrol agents against antibiotic-resistant bacteria.
