Chalcone derivatives have potential as anticancer agents. In this study, furan-based chalcone derivatives were synthesized, and all products were evaluated for cytotoxicity against several cancer cell lines. Chalcones K1 and K2 were synthesized through a Claisen-Schmidt condensation reaction between 2-acetylfuran with 4-dimethylaminobenzaldehyde or 4-chlorobenzaldehyde. Chalcones KA, KB, and KC were synthesized through the same reaction using furfural and 2-chloroacetophenone, 3-chloroacetophenone, or 4-chloroacetophenone. The reaction was carried out by magnetic stirring in methanol, with NaOH as a catalyst. All products were characterized by GC-MS and FTIR spectroscopy. The synthesized compounds were then evaluated for cytotoxic activity using the MTT assay against cervical cancer cells (HeLa), breast (MCF-7 and T47D), colon (WiDr), and normal cells (Vero).

The synthesis of chalcone K1  produced an orange solid with a melting point of 106-108 °C and a yield of 22.82%, while chalcone K2  was obtained as a white solid with a yield of 52.93% and a melting point of 145-147 °C. The synthesis of chalcone KA  produced a dark brown solid with a yield of 84.88% and a melting point of 136-138 °C, chalcone KB produced a light brown solid with a yield of 82.13% and a melting point of 131-133 °C, whereas chalcone  KC was obtained as a white solid with a yield of 91.06% and a melting point of 123-125 °C. All synthesized compounds exhibited high purity. The cytotoxicity assay results showed that the chalcone K2 exhibited the highest cytotoxic activity against breast cancer cells (MCF-7), with an IC50 value of 0.13 ?g/mL and a selectivity index of 120.67. These findings indicate that chalcone K2 has the greatest potential for further investigation as an anticancer agent.
