Roselle (Hibiscus Sabdariffa L.) is a rich source of phenolic compounds with important antioxidant and functional properties. This study evaluated the potential of ATR-FTIR and UV–Vis spectroscopy combined with chemometric approaches for rapid classification of roselle samples and quantitative prediction of major phenolic compounds in roselle powder, using HPLC-PDA as the reference method. Chlorogenic acid, caffeic acid, and rutin were identified and quantified using HPLC-PDA. ATR-FTIR spectra were recorded at 4,000–600 cm?¹, while UV–Vis spectra were obtained at 200–800 nm. Spectral datasets were preprocessed using Standard Normal Variate (SNV), Multiplicative Scatter Correction (MSC), baseline correction, smoothing, and derivative transformations. Sample classification was performed using Support Vector Machine (SVM) and Elastic Net (eNet), while quantitative models were developed using Partial Least Squares Regression (PLSR). The HPLC-PDA method demonstrated excellent analytical performance with high linearity (R2 = 0.994–0.999), low detection limits, and good precision. Cultivar type significantly affected phenolic composition, whereas drying methods showed no significant effect (p > 0.05). ATR-FTIR and UV–Vis spectroscopy achieved high accuracies for cultivar classification (96% and 95%, respectively), although drying-method classification showed moderate accuracies (~80%). In quantitative prediction, UV–Vis spectroscopy demonstrated strong predictive performance for chlorogenic acid (R2P = 0.8858) and superior predictive performance for caffeic acid (R2P = 0.7660), while both techniques showed comparable predictive capability for rutin. External validation confirmed satisfactory predictive performance and model generalizability across independent roselle samples. Overall, ATR-FTIR and UV–Vis spectroscopy demonstrated strong potential as rapid and environmentally sustainable alternatives to conventional HPLC-PDA for routine phenolic analysis and quality assessment of roselle powder.
