Indonesia consist a high abundance of pokea clam populations. Pokea, Batissa violacea (Lamarck, 1818), was one of the freshwater clam species commonly found in the waters of Southeast Sulawesi. However, studies on the genetic variation of pokea clams based on the mitochondrial 16S rRNA gene has been limited in Indonesia. Research on genetic variation was important for constructing a more comprehensive mitochondrial 16S gene database and as a foundation for conservation efforts. Therefore, this study aimed to explore the genetic variation of B. violacea collected from the Konaweha River, Southeast Sulawesi, using the mitochondrial 16S rRNA gene. The method used in this study was Polymerase Chain Reaction (PCR) with universal primers 16Sar and 16Sbr, which included DNA isolation, DNA amplification, electrophoresis, purification, and sequencing. The resulting data were analyzed using GeneStudio, DnaSP, BLAST, MESQUITE, MEGA, NETWORK, BEAST, and GenAlEx software. Based on the results, all samples were successfully identified as Batissa violacea through BLAST analysis, with a similarity value of 98.53% and a query cover of 65% compared to reference data from GenBank. Intrapopulation genetic diversity analysis showed that the pokea population in the Konaweha River had relatively low genetic variation. From ten samples analyzed, 9 haplotypes were identified, with 10 variable site and 2 parsimony informative site. The haplotype diversity value was 0,533 ± 0,095, and the nucleotide diversity (?) was 0,00104 ± 0,00018. Interpopulation analysis between samples from the Konaweha River and comparative data from GenBank showed variation in nucleotide composition and genetic distances ranging from 0.00% to 0.38% among samples and approximately 1.89% to 1.91?tween local samples and the comparative samples. The haplotype diversity value was 0,618 ± 0,104, and the nucleotide diversity (?) was 0,00510 ± 0,00245, with a total of eight haplotypes identified. This study was expected to contribute to the development of a mitochondrial 16S gene database for pokea clams and to serve as a basis for conservation efforts.
