Vanilla (Vanilla planifolia) is a member of the Orchidaceae family that depends on mycorrhizal fungi, particularly Rhizoctonia-like fungi, for seed germination and survival in natural habitats. This study aimed to examine the diversity of macroscopic and microscopic characteristics of Rhizoctonia-like mycorrhizal fungi, identify the isolates using a combination of three gene loci, namely ITS, tef-1, and rpb2, evaluate peloton formation in vitro, and determine the effect of temperature on fungal colony growth. Exploration conducted at 23 different locations yielded four isolates originating from Central Java and Bali. The results showed that the Rhizoctonia-like mycorrhizal fungal isolates exhibited diverse macroscopic characteristics, including colony colours ranging from pale yellow to pale brown, varied colony textures such as waxy flat, velvety, and strip-like forms, different mycelial growth patterns among isolate groups, including non-aerial and slightly aerial mycelia, as well as the presence of concentric rings and sclerotia, which were observed in only one isolate. Microscopically, all isolates showed uniform diagnostic characteristics, including septate hyphae with branching at nearly 90° angles, binucleate cells, and narrow to moderately wide hyphae, although monilioid cells were observed in only two isolates. Incubation temperature significantly affected the growth rate and morphological characteristics of the colonies, with the optimum growth temperature range for the four isolates being 25–28°C. Based on molecular identification using three different primer pairs, namely ITS1/ITS4, EF1-983F/EF1-2218R, and fRPB2-5F/bRPB2-7R2, all four isolates were identified as Tulasnella spp. Furthermore, all isolates were able to form peloton structures in the root cortex tissues of vanilla plantlets under in vitro conditions.
