Latar
belakang:
vitro.
Metode: Penelitian ini merupakan
penelitian eksperimental in vitro menggunakan sel koriokarsinoma (BeWo)
dan sel normal (Vero). Sel diperlakukan dengan hesperetin pada berbagai
konsentrasi.
melalui mekanisme penghambatan fosforilasi.
Kesimpulan: Senyawa hesperetin menunjukkan efek sitotoksik lemah terhadap
sel koriokarsinoma (BeWo) maupun sel normal (Vero). Namun demikian, hesperetin

Background: Choriocarcinoma is a highly aggressive gestational
trophoblastic malignancy with a strong metastatic potential. The
phosphoinositide 3-kinase/protein kinase B (PI3K/AKT) signaling pathway plays a
critical role in the regulation of cell proliferation, survival, and resistance
to apoptosis in cancer cells. Activation of this pathway is characterized by
phosphorylation of AKT at the Ser473 residue. Consequently, inhibition of AKT
phosphorylation has emerged as a potential target for anticancer therapy.
Hesperetin is a naturally occurring flavonoid that has been reported to exhibit
anticancer activity; however, its effects on PI3K/AKT pathway activation in
choriocarcinoma cells remain poorly understood.
Objective: This study aimed to evaluate
the effect of hesperetin on PI3K/AKT pathway activation through inhibition of AKT
phosphorylation at Ser473 in choriocarcinoma cells in vitro.
Methods: This study was conducted as
an in vitro experimental study using choriocarcinoma (BeWo) cells and normal
(Vero) cells. The cells were treated with hesperetin at concentrations ranging from
0 to 300. Cytotoxic effects were evaluated using WST-8 and MTT assays to
determine the IC?? values. The effect of hesperetin on PI3K/AKT pathway
activation was assessed by measuring the levels of AKT phosphorylation at
Ser473 and total AKT expression using Western blot analysis.
Results: Hesperetin reduced BeWo cell viability with IC??
values of 261.59 µM at 24 hours and 241.09 µM at 48 hours. In Vero cells,
higher IC?? values were observed, namely 3180 µM (extrapolated IC??) at 24
hours and 285.28 µM at 48 hours, resulting in selectivity indices of 12.15 (24
hours) and 1.18 (48 hours). Western blot analysis demonstrated that hesperetin
significantly reduced AKT phosphorylation at the Ser473 residue at a
concentration of 125 µM after 48 hours, without affecting total AKT expression,
indicating inhibition of PI3K/AKT pathway activation through suppression of AKT
phosphorylation.
Conclusion: Hesperetin exhibited cytotoxic effects on both
choriocarcinoma (BeWo) cells and normal (Vero) cells. However, hesperetin did
not demonstrate high selectivity toward BeWo cells compared to Vero cells. At a
concentration of 125 µM for 48 hours, hesperetin significantly reduced the
expression of phosphorylated AKT (Ser473) in BeWo cells. In contrast,
hesperetin at concentrations of 150 µM for 24 hours and 125 µM for 48 hours did
not affect the expression of total AKT protein in BeWo cells
