Human papillomavirus (HPV) is a major causative agent of cervical cancer, which remains of the leading cause of cancer related mortality among women in Indonesia. HPV vaccination represent an important strategy for preventing the HPV transmission and reducing the incidence of cervical cancer. However, the high production costs and negative public perception of HPV vaccine remain significant obstacles to the success of vaccination programs. Therefore, efforts are needed to improve the vaccine efficiency and effectiveness vaccines, one of which is through the development of virus-like particle (VLP)-based adjuvants capable of enhancing the immune response thereby reducing antigen dose requirements and overall production costs. This study aimed to evaluate the potential of chitosan-propolis as a nanoparticle candidate adjuvant for VLP-based vaccines through in vivo studies. The study was conducted with several stages including formulation optimization, stability testing, to animal experiment. In vivo testing was conducted for 42 days using mice (Mus musculus Linnaeus, 1758). The test animals were divided into 4 groups, namely the negative control group without adjuvant administration without VLP, the adjuvant administration group without VLP, the VLP administration group without adjuvant, and the NKP-VLP HPV vaccine preformula administration group. Characterization analysis, demonstrated the particle size, particle size distribution, and zeta potential values for the formation of stable nanoparticle system with an optimal encapsulation efficiency 91.371 ± 0.007%.
