Formulasi Nanostructured Lipid Carriers Terliofilisasi Memuat Fraksi Kaya Alfa-Mangostin Kulit Buah Manggis (Garcinia mangostana L.) untuk Peningkatan Penghantaran Transdermal

Alfa-mangostin merupakan senyawa xanthone mayor dalam kulit buah Garcinia mangostana L. yang diklasifikasikan sebagai biopharmaceutical classification system (BCS) kelas II dengan bioavailabilitas oral yang terbatas. Penghantaran transdermal menggunakan nanostructured lipid carriers (NLC) menawarkan strategi yang menjanjikan untuk mengatasi keterbatasan ini. Penelitian ini bertujuan untuk memformulasikan sistem NLC yang menggabungkan fraksi kaya alfa-mangostin (FKAM) untuk meningkatkan kinerja penghantaran transdermal. FKAM diperoleh melalui ultrasonic-assisted extraction menggunakan pelarut metanol diikuti dengan fraksinasi etil asetat, dan senyawa alfa-mangostin dikuantifikasi secara HPTLC, HPLC, dan spektrofotometri UV-Vis. NLC diformulasikan menggunakan glyceryl monostearate (GMS), minyak kelapa (CO), dan surfaktan mix (Smix) yang tersusun atas Polysorbate 20 (P20) dan Sorbitan Laurate (SL) dengan HLB 10. Metode D-Optimal Mixture Design digunakan untuk mengoptimasi parameter ukuran partikel, indeks polidispersitas, zeta potensial, dan efisiensi enkapsulasi. NLC optimal diliofilisasi dengan trehalose dan diredispersi berbantu ultrasonic kemudian dikarakterisasi menggunakan PSA, TEM, SEM, serta dilakukan uji pelepasan in vitro dan permeasi ex vivo. Formulasi NLC optimal dengan komposisi GMS (3%b/b), CO (1,466%b/b), dan Smix (5,534%b/b) menghasilkan ukuran partikel (181,02 ±10,579 nm), PDI (0,367 ± 0,039), potensial zeta negatif (-32,04 ± 1,201 mV), dan efisiensi enkapsulasi (98,859 ± 0,041). Validasi HLB Smix mengkonfirmasi bahwa surfaktan mix HLB 10 menghasilkan sistem yang paling sesuai dengan kriteria. NLC terliofilisasi dengan 5% trehalose dan durasi 5 menit sonikasi menunjukkan redispersabilitas yang baik dengan karakteristik ukuran partikel (198,97 ± 1,155 nm), PDI (0,268±0,005), dan zeta potensial (?28,267±0,493 mV). Pelepasan in vitro mengikuti model kinetika Korsmeyer-Peppas dengan difusi anomali dan pelepasan berkelanjutan dibandingkan dengan kontrol FKAM. Studi permeasi ex vivo menggunakan kulit tikus menunjukkan peningkatan 2,15 kali lipat dalam permeasi kumulatif dan fluks transdermal yang lebih tinggi. Hal ini dikaitkan dengan ukuran skala nano, interaksi lipid-kulit, dan transport via folikel. NLC-FKAM optimal secara signifikan meningkatkan penghantaran transdermal alfa-mangostin, sehingga mendukung aplikasi potensialnya dalam aplikasi transdermal untuk senyawa bioaktif lipofilik dengan baik.
Alpha-mangostin, the principal xanthone in Garcinia mangostana L., is classified as a biopharmaceutics classification system (BCS) class II compound with limited oral bioavailability. Transdermal delivery using nanostructured lipid carriers (NLC) offers a promising strategy to overcome this limitation. In this study, an NLC system incorporating an alpha-mangostin-rich fraction (AMRF) was developed to enhance transdermal delivery. AMRF was extracted by ultrasonic-assisted extraction with methanol, followed by ethyl acetate fractionation, and quantified using HPTLC, HPLC, and UV-Vis spectrophotometry. NLCs were prepared with glyceryl monostearate (GMS), coconut oil (CO), and a surfactant mixture (Smix) of Polysorbate 20 and Sorbitan Laurate. The D-Optimal Mixture Design method was used to optimize particle size, polydispersity index (PDI), zeta potential, and encapsulation efficiency. Optimized NLCs were lyophilized with trehalose and characterized by particle size analysis, transmission electron microscopy (TEM), and scanning electron microscopy (SEM). In vitro release and ex vivo skin permeation studies were conducted. The optimal NLC with GMS (3% w/w), CO (1,466% w/w), and Smix (5,534% w/w) exhibited a particle size of 181.02 ± 10.579 nm, a PDI of 0.367 ± 0.039, a zeta potential of -32.04 ± 1.201 mV, and an encapsulation efficiency of 98.859 ± 0.041%. Hydrophilic-lipophilic balance (HLB) validation showed that a surfactant HLB of 10 produced the most suitable criteria system. NLCs lyophilized with 5% trehalose and sonicated for 5 minutes demonstrated favorable redispersibility with particle size (198,97 ± 1,155 nm), PDI (0,268±0,005), and zeta potential (?28,267±0,493 mV). In vitro release studies followed the Korsmeyer-Peppas kinetic model, indicating anomalous diffusion and sustained release compared to the AMRF control. Ex vivo permeation studies using rat skin demonstrated a 2.15-fold increase in cumulative permeation and higher transdermal flux, attributed to nanoscale particle size, lipid-skin interactions, and follicular transport. These results indicate that the optimized AMRF-NLC formulation significantly improves the transdermal delivery of alpha-mangostin and supports its potential application for the transdermal administration of lipophilic bioactive compounds.
