SIFAT ANTIMIKROBA DAN LAJU PELEPASAN MINYAK CENGKEH PADA PENGEMASAN AKTIF KITOSAN – MIKROKRISTALIN SELULOSA

Penelitian
ini mengevaluasi karakteristik, pelepasan eugenol, dan efek antimikroba dari
film komposit kitosan/mikrokristalin selulosa yang mengandung minyak cengkeh
pada kontak langsung dan tidak langsung. Efek rasio kitosan terhadap
mikrokristalin selulosa (MCC) dan kandungan minyak cengkeh terhadap
karakteristik kimia, morfologi, fisik, serta karakteristik berbasis air dari
film komposit. Pelepasan senyawa dinilai dengan mengukur konsentrasi eugenol
dan efek antimikrobanya terhadap Escherichia coli, Staphylococcus aureus,
dan Bacillus
subtilis. Penambahan rasio MCC menghasilkan
permukaan film yang bertekstur dan berpori, lebih tebal, dan lebih buram,
tetapi menurunkan sifat berbasis air. Sementara itu, penambahan minyak cengkeh
menghasilkan film yang lebih tebal dan lebih buram, meningkatkan pembengkakan
dan transmisi air, tetapi menurunkan hidrofobisitas. Pada sistem kontak
langsung, konsentrasi MCC tidak memberikan pengaruh signifikan terhadap
pelepasan senyawa eugenol tetapi menunjukkan efek yang jelas dari konsentrasi
minyak cengkeh yang menghasilkan pelepasan eugenol lebih tinggi seiring
peningkatan kandungan minyak cengkeh. Minyak cengkeh efektif dalam menghambat
pertumbuhan bakteri pada sistem pengemasan kontak langsung tetapi tidak
menunjukkan dampak ketika dievaluasi dengan metode kontak tidak langsung karena
konsentrasi eugenol headspace yang rendah. Oleh karena itu, film
yang dikembangkan lebih aplikatif pada sistem pengemasan pangan kontak
langsung.
This
study evaluated characteristics, eugenol release and antimicrobial effects of a
chitosan/microcrystalline cellulose composite film containing clove oil under
direct and indirect contact. The effects of ratios of chitosan to
microcrystalline cellulose (MCC) and the clove oil content on the chemical,
morphological, physical, water-based characteristics of the film were
investigated. Compound release was assessed by measuring eugenol concentration
and its antimicrobial effects against Escherichia coli, Staphylococcus
aureus, and Bacillus subtilis. The addition of MCC ratios produced
film surfaces that were textured and porous, thicker, and opaquer, but reduced
water based properties. Meanwhile, the addition of clove oil produced films
that were thicker and more opaque, increased swelling and water transmission,
but decreased hydrophobicity. In the direct contact system, MCC concentration
did not exert a significant effect on the release of eugenol compounds but
showed a clear effects of clove oil concentration producing a higher eugenol
release in an increase of clove oil content. The incorporation of clove oil was
effective in inhibiting bacterial growth under direct-contact packaging system
but showed no impact when evaluated under indirect contact methods due to low
headspace concentration observed. Therefore, the developed film is more
applicable in direct food packaging system.
