ISOLASI DAN KARAKTERISASI BAKTERIOFAG PENGENDALI Vibrio parahaemolyticus DARI AIR TAMBAK UDANG VANAME DI KABUPATEN PURWOREJO

Penelitian ini bertujuan untuk mendapatkan isolat bakteriofag dari air tambak udang vaname yang diketahui karakter dan potensinya untuk mengendalikan infeksi Vibrio parahaemolyticus. Penelitian dilakukan dengan cara mengisolasi bakteriofag menggunakan metode plaque assay pada media Zobell dan karakterisasi dengan pengamatan morfologi menggunakan transmission electron microscopy (TEM), uji kemampuan produksi, uji rentang inang (Host range), uji kemampuan in vitro pengendalian pertumbuhan V. parahaemolyticus BKIPM dan uji in vivo pengendalian infeksi V. parahaemolyticus pada udang vaname. Fag PUF25 pengendali V. parahaemolyticus berhasil diisolasi dari air tambak udang vaname  dengan kemampuan produksi sebesar 1,24 x 106 PFU/ml. Fag PUF25 memiliki morfologi menyerupai famili Podoviridae berukuran 64,8 ± 23,7 nm dengan kepala  ikosahedral (50,4 ± 20,8 nm) dan ekor pendek (14,4 ± 4,4 nm). Fag PUF25 memiliki spektrum inang 33%, yaitu  menghambat V. parahaemolyticus BKIPM, V. parahaemolyticus CVB dan Photobacterium damselae subsp damselae GD05, dan tidak menghambat enam strain Vibrio lainnya. This study aims to obtain bacteriophage isolates from whiteleg shrimp pond water whose characteristics and potential are known to control Vibrio parahaemolyticus infection. The study was conducted by isolating bacteriophages using the plaque assay method on Zobell media and characterizing them by morphological observation using transmission electron microscopy (TEM), production capability test, host range test, in vitro capability test to control V. parahaemolyticus BKIPM growth and in vivo test to control V. parahaemolyticus infection in whiteleg shrimp. The PUF25 phage controlling V. parahaemolyticus was successfully isolated from whiteleg shrimp pond water with a production capability of 1.24 x 106 PFU/ml. The PUF25 phage has a morphology resembling the Podoviridae family measuring 64,8 ± 23,7 nm with an icosahedral head (50,4 ± 20,8 nm) and a short tail (14,4 ± 4,4 nm). The PUF25 phage has a 33% host spectrum, which inhibits V. parahaemolyticus BKIPM, V. parahaemolyticus CVB and Photobacterium damselae subsp damselae GD05, and does not inhibit six other Vibrio strains.
