2026 | Skripsi | BIOLOGINuri Talaud (Eos histrio) merupakan spesies burung endemik Pulau Karakelang yang mengalami penurunan drastis yang signifikan populasinya akibat aktivitas perburuan, perdagangan ilegal, serta degradasi habitat. Eos histrio dikategorikan sebagai terancam punah (Endangered/EN) oleh IUCN dan dilindungi berdasarkan Peraturan Menteri Lingkungan Hidup dan Kehutanan Nomor 106 Tahun 2018. Konservasi genetik memegang peranan penting dalam upaya pelestarian Eos histrio, sekaligus informasi pendukung tentang biogeografi dan filogenetik historis pada spesies ini, yang hingga saat ini masih minim tersedia informasi genetiknya. Penelitian ini bertujuan untuk menganalisis filogenetik dan biogeografi Eos histrio menggunakan fragmen DNA mitokondria Cytochrome b guna memahami sejarah evolusi dan pola biogeografi. Sampel DNA diperoleh dari individu yang diselamatkan di Pusat Penyelamatan Satwa Tasikoki, Bitung, Sulawesi Utara. Metode penelitian ini meliputi ekstraksi DNA, Polymerase Chain Reaction (PCR), serta elektroforesis untuk verifikasi keberhasilan amplifikasi. Produk PCR selanjutnya dikirim untuk sekuensing dan dianalisis data sekuensnya. Rekonstruksi filogenetik dilakukan menggunakan model Bayesian inference. Titik-titik kalibrasi digunakan untuk memperkirakan waktu divergensi pada Eos histrio berdasarkan pohon Filogenetik Bayesian. Estimasi wilayah leluhur pada nenek moyang Eos histrio menggunakan Reconstruct Ancestral State in Phylogenies dilakukan untuk menguji rute dispersal dan vikarian Eos histrio. Pohon filogenetik mengungkap Eos histrio bercabang selama Pleistocene Tengah 0.92 juta tahun yang lalu (interval HPD: 0.2–1,63 juta tahun yang lalu). Rekonstruksi pohon filogenetik mengungkapkan rute dispersal dan vikarian yang terjadi pada Eos histrio, yang berawal dari Papua, Maluku, Sulawesi Tengah, Sulawesi Utara, stepping-stone dispersal ke Pulau Sangihe baru berakhir di Pulau Karakelang.
The Red and Blue Lory (Eos histrio) is an endemic bird species of Karakelang Island that has experienced a drastic population decline due to hunting activities, illegal trade, and habitat degradation. Eos histrio is categorized as Endangered (EN) by the IUCN Red List and is protected under the Minister of Environment and Forestry Regulation Number 106 of 2018. Genetic conservation contributes significantly to the conservation of Eos histrio by providing critical data on its biogeography and evolutionary history, particularly given the current scarcity of genetic information for this species. This study aims to analyze the phylogenetic relationships of Eos histrio using mitochondrial DNA Cytochrome b fragment to understand the evolutionary history and biogeographic patterns of the species. DNA samples were obtained from individuals rescued at the Tasikoki Wildlife Rescue Center, Bitung, North Sulawesi. The research methods included DNA extraction, Polymerase Chain Reaction (PCR) amplification, and electrophoresis for verification of amplification success. PCR products were subsequently sent for sequencing, and sequence data were analyzed using Bayesian inference model. Calibration points were used to estimate divergence time of Eos histrio based on a Bayesian phylogenetic tree. Ancestral area reconstruction using the Reconstruct Ancestral State in Phylogenies (RASP) method was conducted to examine the dispersal routes and vicariance events of Eos histrio. The phylogenetic tree revealed that Eos histrio diverged during the Middle Pleistocene, approximately 0.92 Ma (highest posterior density interval: 0.2–1.63 million years ago). Phylogenetic reconstruction revealed the dispersal routes and vicariance events occurring in Eos histrio, originating from Papua, Maluku, Central Sulawesi, North Sulawesi, with stepping-stone dispersal to Sangihe Island, and finally ending in Karakelang Island.
