Banana bunchy top virus (BBTV) merupakan salah satu patogen penting pada tanaman pisang karena dapat menyebabkan kerugian hingga 100% akibat kegagalan pembentukan buah. Penelitian ini bertujuan untuk mengidentifikasi keberadaan BBTV secara molekuler pada beberapa tanaman inang, menganalisis kekerabatan genetik isolat yang diperoleh dengan isolat lain di NCBI GenBank, dan mengkaji potensi inang alternatif bagi BBTV di wilayah Daerah Istimewa Yogyakarta dan Jawa Tengah. Deteksi molekuler dilakukan melalui ekstraksi DNA, amplifikasi menggunakan teknik Polymerase Chain Reaction (PCR) dengan primer spesifik DNA-R, serta visualisasi hasil menggunakan elektroforesis gel agarosa. Analisis sekuens dilakukan dengan perangkat lunak MEGA untuk menyusun hubungan filogenetik dengan metode bootstrap 1000 kali serta analisis matriks pairwise identity. Hasil penelitian menunjukkan bahwa BBTV tidak hanya terdeteksi pada tanaman pisang, tetapi juga menginfeksi tanaman lain sebagai inang alternatif dari genus Zingiberaceae dan Cannaceae. Analisis filogenetik menunjukkan adanya kedekatan genetik antar isolat lokal dengan isolat dari wilayah lain, yang mengindikasikan adanya penyebaran virus melalui perbanyakan tanaman dan serangga vektor. Keberadaan inang alternatif berperan penting sebagai reservoir virus dan dapat mempertahankan keberlanjutan infeksi penyakit di lapangan. Penelitian ini memberikan informasi penting mengenai keragaman genetik dan dinamika penyebaran BBTV, serta dapat menjadi dasar dalam penyusunan strategi pengendalian penyakit yang lebih efektif melalui pengelolaan bahan tanam, sanitasi lahan, pengendalian vektor, dan terutama pada pengelolaan tanaman inang alternatif.
Banana bunchy top virus (BBTV) is one of the major pathogens affecting banana plants, as it can cause up to 100% yield loss due to failure in fruit set. This study aims to identify the presence of BBTV molecularly in several host plants, analyze the genetic relationship of the obtained isolates with other isolates in the NCBI GenBank, and assess the potential of alternative hosts for BBTV in the Special Region of Yogyakarta and Central Java. Molecular detection was performed through DNA extraction, amplification using the Polymerase Chain Reaction (PCR) technique with DNA-R-specific primers, and visualization of the results using agarose gel electrophoresis. Sequence analysis was conducted using MEGA software to construct a phylogenetic tree using the 1000-bootstrap method and to analyze pairwise identity matrix. The results of the study indicate that BBTV is not only detected in banana plants but also infects other plants as alternative hosts from the genera Zingiberaceae and Cannaceae. Phylogenetic analysis revealed genetic proximity between local isolates and those from other regions, indicating virus spread through plant propagation and insect vectors. The presence of alternative hosts plays a crucial role as a viral reservoir and can sustain disease infection in the field. This study provides important information on the genetic diversity and spread dynamics of BBTV and can serve as a basis for developing more effective disease control strategies through the management of planting materials, field sanitation, vector control, and, in particular, the management of alternative host plants.
