Penelitian ini bertujuan untuk mengetahui produksi
terbaik enzim fibrolitik dari isolat campuran bakteri rumen kerbau pada variasi
substrat berupa kapas, ampas jagung, dan jerami padi, serta mengetahui
aktivitas enzim optimum pada pH 4, 5, 6, 7, dan 8. Isolat bakteri yang
digunakan berasal dari rumen kerbau yang diliofilisasi (freeze dried) koleksi
Lab. Biokimia Nutrisi, Fakultas Peternakan, UGM. Produksi enzim dilakukan
dengan mengkultur isolat pada medium Hungate dan diinkubasi selama 7 hari menggunakan
ketiga jenis substrat pada level 0,5% untuk mencari substrat terbaik. Substrat
terbaik digunakan lebih lanjut untuk produksi enzim pada karakterisasi pH. Replikasi
kultur dilakukan sebanyak empat kali. Ekstraksi enzim dilakukan dengan proses
sentrifugasi, kemudian supernatan digunakan sebagai sumber enzim. Parameter

This study aimed to determine the optimal production of fibrolytic enzymes from a mixed isolate of buffalo rumen bacteria using various substrates, including cotton, corn cob, and rice straw, as well as to determine the optimal enzyme activity at pH levels of 4, 5, 6, 7, and 8. The bacterial isolates used were derived from freeze-dried buffalo rumen samples from the collection of the Laboratory of Nutritional Biochemistry, Faculty of Animal Science, UGM. Enzyme production was carried out by culturing the isolates on Hungate medium and incubating them for 7 days using all three substrate types at 0.5% concentration to identify the optimal substrate. The best substrate was further used for enzyme production in the pH characterization. Culture replicates were performed four times. Enzyme extraction was carried out via centrifugation, and the supernatant was used as the enzyme source. The study parameters were the activities of endoglucanase, ?-glucosidase, exoglucanase, and xylanase. Data were analyzed using one-way analysis of variance (ANOVA) following a completely randomized design (CRD). If differences were found among treatments, the analysis was continued with Duncan’s Multiple Range Test (DMRT).
