Pengaruh Variasi HPMC dan CMC-Na terhadap Stabilitas dan Sifat Fisik Serum Tetrahidropentagamavunon-0 (THPGV-0)

Paparan radiasi matahari dan polusi udara berlebih dapat memicu  peningkatan produksi radikal bebas sehingga mempercepat penuaan kulit. Senyawa Tetrahidropentagamavunon-0 (THPGV-0) memiliki aktivitas antioksidan yang berguna untuk menetralkan radikal bebas. Aktivitas antioksidan THPGV-0 yang tinggi dapat dimanfaatkan sebagai kosmetik antiaging seperti serum. Serum memiliki konsentrasi zat aktif tinggi dan viskositas rendah. Viskositas serum yang baik dapat diperoleh menggunakan thickening agent yang sesuai. Penelitian ini bertujuan untuk mengoptimasi thickening agent kombinasi HPMC dan CMC-Na untuk mendapatkan serum dengan stabilitas dan sifat fisik yang optimal, serta mengetahui aktivitas antioksidan serum THPGV-0.

Penelitian diawali dengan optimasi formula basis serum menggunakan metode Simplex Lattice Design pada software Design Expert. Selanjutnya, dilaksanakan pembuatan serum menggunakan formula optimum dan diuji stabilitas fisiknya menggunakan metode cycling test selama tiga siklus. Data dianalisis menggunakan software IBM SPSS Statistic dengan taraf kepercayaan 95%. Selain itu, aktivitas antioksidan senyawa THPGV-0 dan serum THPGV-0 diuji menggunakan metode DPPH. 

Hasil penelitian menunjukkan formula optimum basis serum memiliki kombinasi HPMC 0,45?n CMC-Na 0,55%. Respons sifat fisik yang diperoleh, yaitu viskositas 239,30±2,83 cP, pH 5,86±0,02, dan diameter sebar 8,78±0,01 cm. Serum THPGV-0 bersifat stabil pada respons viskositas, pH, dan diameter sebar selama tiga siklus. Namun, mengalami perubahan pada respons organoleptis dan homogenitas yang ditandai dengan adanya endapan. Selain itu, diperoleh nilai IC50 senyawa THPGV-0 dan serum THPGV-0 secara berturut-turut sebesar 22,03 ?M dan 21,64 ?M.

Excessive exposure to sunlight and air pollution can trigger an increase in free radical production, thereby accelerating skin aging. The compound Tetrahydropentagamavunon-0 (THPGV-0) possesses antioxidant activity that is useful for neutralizing free radicals. The high antioxidant activity of THPGV-0 can be utilized in antiaging cosmetics such as serums. Serums have a high concentration of active ingredients and low viscosity. Optimal serum viscosity can be achieved using an appropriate thickening agent. This study aimed to optimize a combination of HPMC and CMC-Na as thickening agents to obtain a serum with optimal stability and physical properties, as well as to determine the antioxidant activity of THPGV-0 serum.

The study began with optimization of the serum base formula using the Simplex Lattice Design method in Design Expert software. Subsequently, the serum was prepared using the optimized formula and its physical stability was evaluated using a cycling test for three cycles. The data were analyzed using IBM SPSS Statistics software with a 95% confidence level. Additionally, the antioxidant activity of the THGPV-0 compound and THGPV-0 serum was tested using the DPPH method.

The results showed that the optimum serum base formula had a combination of 0.45% HPMC and 0.55% CMC-Na. The physical properties obtained were viscosity of 239.30±2.83 cP, pH 5.86±0.02, and spreadability of 8.78±0.01 cm. THPGV-0 serum was stable in terms of viscosity, pH, and spreadability over three cycles. However, changes were observed in the organoleptic properties and homogeneity, indicated by the presence of sediment. Furthermore, the IC50 values for the THPGV-0 compound and THPGV-0 serum were 22.03 ?M and 21.64 ?M, respectively.
