Evaluasi Eksopolisakarida Lactobacillus bulgaricus sebagai Agen Biopreservasi pada Ikan Tuna selama Masa Penyimpanan

Ikan merupakan sumber pangan dengan nilai gizi yang tinggi. Namun, produk ikan merupakan bahan pangan yang mudah rusak karena kandungan air yang tinggi, pH netral, dan kandungan gizi yang mendukung pertumbuhan bakteri pembusuk sehingga biasanya digunakan pengawet sintetis untuk memperpanjang masa simpan. Akan tetapi, pengawet sintetis memiliki risiko kesehatan yang signifikan, termasuk potensi efek karsinogenik dan sitotoksik. Sebagai alternatif eksopolisakarida (EPS) yang dihasilkan oleh Bakteri Asam Laktat Lactobacillus bulgaricus mampu menghambat pertumbuhan bakteri patogen terbawa pangan melalui mekanisme edible coating. Penelitian ini bertujuan untuk mengevaluasi efektivitas eksopolisakarida yang dihasilkan oleh Lactobacillus bulgaricus dalam mengawetkan produk pangan ikan selama masa penyimpanan. Penelitian ini akan melibatkan beberapa metode termasuk kultur Lactobacillus bulgaricus dan ekstraksi eksopolisakarida dari Lactobacillus bulgaricus dengan metode presipitasi protein dengan etanol, pembuatan edible coating berbasis eksopolisakarida dengan dua variasi konsentrasi yaitu EPS 0,5?n EPS 1%. Parameter yang diuji selama masa penyimpanan pada suhu 4? meliputi analisis proksimat, analisis TVB-N, nilai pH, analisis angka lempeng total, deteksi E. coli dan Salmonella sp. serta uji sensori. Hasil penelitian menunjukkan bahwa proses ekstraksi menghasilkan rendemen eksopolisakarida kasar yang signifikan sebesar 7,84%. Aplikasi EPS sebagai edible coating secara nyata mampu mempertahankan profil nutrisi makro ikan tuna hingga hari ke-3 penyimpanan dengan mempertahankan kadar air dibanding dengan kontrol negatif aquades. Selain itu, perlakuan EPS 0,5% mampu menahan laju kenaikan pH dengan nilai rata-rata 5,88 dan menghambat proliferasi mikroba yang ditunjukkan hasil TPC pada hari ke-6 sebesar 15,5 x 102 CFU/g dibandingkan dengan kontrol positif (Natrium Benzoat) yang memiliki nilai 30,4 x 102 CFU/g. Secara sensori, penggunaan EPS belum mampu mempertahankan bau dan tekstur produk lebih lama dibandingkan kontrol poisitif (natrium benzoat). Kesimpulannya, eksopolisakarida dari L. bulgaricus memiliki potensi sebagai agen pengawet alami yang efektif dalam memperpanjang masa simpan dan menjaga kualitas fisikokimia serta organoleptik ikan tuna.

Fish is a food source with high nutritional value. However, fish products are highly perishable due to their high water content, neutral pH, and nutrient composition that supports the growth of spoilage bacteria; therefore, synthetic preservatives are typically used to extend their shelf life. However, synthetic preservatives pose significant health risks, including potential carcinogenic and cytotoxic effects. As an alternative, exopolysaccharides (EPS) produced by the lactic acid bacterium Lactobacillus bulgaricus can inhibit the growth of foodborne pathogenic bacteria through an edible coating mechanism. This study aims to evaluate the effectiveness of exopolysaccharides produced by Lactobacillus bulgaricus in preserving fish products during storage. This study will involve several methods, including the culture of Lactobacillus bulgaricus and the extraction of exopolysaccharides from Lactobacillus bulgaricus using the ethanol protein precipitation method, as well as the preparation of an exopolysaccharide-based edible coating with two concentration variations: 0,5% EPS and 1% EPS. Parameters tested during storage at 4°C included proximate analysis, TVB-N analysis, pH value, total plate count analysis, detection of E. coli and Salmonella sp., and sensory evaluation. The results showed that the extraction process yielded a significant crude exopolysaccharide yield of 7,84%. The application of EPS as an edible coating was clearly able to maintain the macro-nutritional profile of tuna through the third day of storage by maintaining moisture content compared to the distilled water negative control. Additionally, the 0,5% EPS treatment was able to inhibit the rate of pH increase, with an average value of 5,88, and suppress microbial proliferation, as evidenced by the TPC result on day 6 of 15,5 × 10² CFU/g compared to the positive control (sodium benzoate), which had a value of 30,4 × 10² CFU/g. Sensory evaluation revealed that the use of EPS was not yet able to maintain the odor and texture of the product for a longer period compared to the positive control (sodium benzoate). In conclusion, exopolysaccharides from L. bulgaricus have the potential as an effective natural preservative in extending the shelf life and maintaining the physicochemical and organoleptic quality of tuna.
