Uji Efek Hepatoprotektif Andrographis paniculata terhadap Parameter Kimia Darah Serum Glutamic Pyruvic Transaminase dan Gambaran Makroskopis Hepar Mencit yang Diinduksi Parasetamol secara Per Oral

Parasetamol merupakan obat analgesik dan antipiretik yang banyak digunakan untuk meredakan nyeri dan menurunkan demam. Penggunaan parasetamol dalam dosis toksik dapat menyebabkan hepatotoksisitas akibat pembentukan metabolit reaktif yang memicu stres oksidatif, inflamasi, dan nekrosis sel hepatosit. Tanaman herbal yang berpotensi memiliki efek hepatoprotektif adalah sambiloto (Andrographis paniculata) yang mengandung senyawa aktif andrographolide. Tugas Akhir ini bertujuan untuk mengetahui efek hepatoprotektif ekstrak sambiloto terhadap kadar Serum Glutamic Pyruvic Transaminase (SGPT) dan gambaran makroskopis hepar mencit (Mus musculus) yang diinduksi parasetamol dosis toksik secara per oral. Penelitian ini merupakan penelitian eksperimental laboratorik rancangan acak lengkap (RAL). Sebanyak 25 ekor mencit jantan sebagai hewan coba dibagi ke dalam lima kelompok sebagai replikasi biologis. Kelompok kontrol negatif (K-) tidak diberi perlakuan; kelompok kontrol positif (K+) diinduksi parasetamol dosis toksik tanpa pemberian ekstrak sambiloto; kelompok perlakuan P1, P2, dan P3 masing-masing diberi ekstrak sambiloto dosis 50, 100, dan 200 mg/kg BB serta diinduksi parasetamol dosis toksik. Perlakuan diberikan selama 15 hari. Enam jam setelah perlakuan hari terakhir, dilakukan induksi parasetamol pada kelompok K+, P1, P2, dan P3. Sampel darah diambil 18 jam setelah induksi untuk pemeriksaan kadar SGPT, kemudian dilakukan nekropsi untuk mengamati gambaran makroskopis hepar meliputi warna, permukaan, konsistensi, dan sudut. Analisis kadar SGPT diawali dengan uji normalitas Shapiro-Wilk dan pemeriksaan grafik Q-Q plot yang menunjukkan data tidak terdistribusi normal (p > 0,05). Hasil analisis statistik menunjukkan pemberian ekstrak sambiloto dosis 50, 100, dan 200 mg/kg BB selama 15 hari belum menunjukkan efek hepatoprotektif terhadap kadar SGPT maupun gambaran makroskopis hepar pada mencit yang diinduksi parasetamol dosis toksik secara per oral.
Paracetamol is an analgesic and antipyretic drug widely used to relieve pain and reduce fever. The use of paracetamol at toxic doses can cause hepatotoxicity due to the formation of reactive metabolites that trigger oxidative stress, inflammation, and hepatocyte necrosis. One herbal plant with potential hepatoprotective effects is sambiloto (Andrographis paniculata), which contains the active compound andrographolide. This final project aims to determine the hepatoprotective effect of sambiloto extract on Serum Glutamic Pyruvic Transaminase (SGPT) levels and the macroscopic appearance of the liver in mice (Mus musculus) induced with toxic doses of paracetamol administered orally. This experimental laboratory study used a completely randomized design (CRD). A total of 25 male mice were divided into five groups as biological replicates. The negative control group (K-) received no treatment; the positive control group (K+) was induced with a toxic dose of paracetamol without administration of sambiloto extract; while the treatment groups P1, P2, and P3 received sambiloto extract at doses of 50, 100, and 200 mg/kg body weight, respectively, and were also induced with a toxic dose of paracetamol. The treatment was administered for 15 days. Six hours after the last day of treatment, paracetamol induction was performed in groups K+, P1, P2, and P3. Blood samples were collected 18 hours after induction for SGPT examination, followed by necropsy to observe the macroscopic appearance of the liver including color, surface, consistency, and edges. SGPT data were analyzed using the Shapiro–Wilk normality test and Q-Q plot, which indicated non-normal distribution (p > 0,05). These results indicate that sambiloto extract at doses of 50, 100, and 200 mg/kg body weight for 15 days did not show hepatoprotective effects.
