Intisari

Indonesia merupakan negara dengan keanekaragaman tanaman yang tinggi, termasuk diantaranya tanaman obat. Secara etnobotani, tanaman obat memiliki banyak peran untuk kesehatan, salah satunya sebagai antibakteri. Kremah (Alternanthera philoxeroides) menjadi salah satu tanaman yang diketahui memiliki sifat antibakteri karena mengandung senyawa metabolit sekunder seperti flavonoid, alkaloid, tannin, saponin, terpenoid, steroid, dan glikosida. Biosintesis senyawa metabolit tanaman tergantung pada kondisi lingkungan tempat tumbuh yang berimplikasi pada efektivitas antibakterinya. Tujuan dari penelitian ini adalah untuk mengetahui perbandingan komposisi kandungan metabolit sekunder dan aktivitas antibakteri tumbuhan A.philoxeroides yang tumbuh di dataran tinggi Kejajar (Wonosobo, Jawa Tengah) dan dataran rendah Ngemplak (Sleman, Yogyakarta). Pengujian dilakukan dengan mengekstraksi tanaman dengan maserasi bertingkat kloroform-etanol 70%, deteksi kualitatif fitokimia dengan reagen, uji kadar flavonoid dengan metode kolorimetri, uji aktivitas antibakteri ekstrak dengan difusi cakram dan MIC, serta eksplorasi senyawa dengan GC-MS. Tumbuhan kremah yang tumbuh di kedua lokasi memiliki kesamaan komposisi golongan senyawa. Kadar flavonoid, saponin, tanin, fenol, dan glikosida relatif lebih tinggi di Kejajar, sedangkan untuk kadar relatif senyawa steroid dan terpenoid lebih tinggi di Ngemplak. Kadar flavonoid ekstrak Kejajar lebih tinggi dibandingkan Ngemplak, yakni 16,16 mgQE/g (kloroform) dan 12,05 mgQE/g (etanol 70%). Aktivitas antibakteri tertinggi ditunjukkan oleh ekstrak kloroform Ngemplak dengan rerata diameter zona hambat sebesar 4.42 mm (Staphylococcus aureus) dan 2.50 mm (Escherichia coli) dengan kategori lemah, serta inhibisi S.aureus terlihat pada konsentrasi 2.500 µg/ml sebagai nilai MIC. Ekstrak tersebut relatif didominasi kelompok senyawa asam lemak, terpenoid, dan sterol yang terdeteksi dalam uji GC-MS.

Abstract

Indonesia is a country with high plant diversity, including medicinal plants. Ethnobotanically, medicinal plants have many roles for health, one of which is as an antibacterial. Kremah (Alternanthera philoxeroides) is one of the plants known as antibacterial properties because it contains secondary metabolite compounds such as flavonoids, alkaloids, tannins, saponins, terpenoids, steroids, and glycosides. The biosynthesis of plant metabolite compounds depends on the environmental conditions where it grows, which has implications for its antibacterial effectiveness. This research aims to compare the composition of secondary metabolite content and antibacterial activity of A.philoxeroides plants growing in the Kejajar highlands (Wonosobo, Jawa Tengah) and the Ngemplak lowlands (Sleman, Yogyakarta). Testing was carried out by extracting plants with chloroform-ethanol 70% multistep maceration, qualitative phytochemical screening with reagents, flavonoid content by colorimetric method, antibacterial activity by disc diffusion and MIC, and compound exploration using GC-MS. Kremah plants from both locations has similar composition compound groups. Level relative of flavonoid, saponin, tanin, phenol, and glycoside are higher in Kejajar, while steroid and terpenoid level relative are higher in Ngemplak. Flavonoid content of Kejajar extract was higher than Ngemplak, with total of 16.16 mgQE/g (chloroform) and 12.05 mgQE/g (ethanol 70%). The highest antibacterial activity was shown by the chloroform extract of Ngemplak with an average inhibition zone diameter of 4.42 mm (Staphylococcus aureus) and 2.50 mm (Escherichia coli) with a weak category, and S.aureus inhibition was seen at 10,000 µg/ml as the MIC value. The extract was relatively dominated by the fatty acid, terpenoid, and sterol compound groups in the GC-MS test.
