Kasus gigitan ular berbisa termasuk ke dalam salah satu masalah kesehatan yang cukup serius di berbagai negara, terutama di negara dengan iklim tropis seperti Asia Tenggara, termasuk Indonesia. Berdasarkan data oleh World Health Organization pada tahun 2024, diperkirakan kasus gigitan ular telah menyebabkan sekitar 81.000 hingga 138.000 kematian dengan 400.000 kasus mengalami kecacatan permanen di seluruh dunia. Salah satu spesies ular berbisa yang dapat ditemukan cukup banyak di Indonesia yaitu ular Trimeresurus insularis atau dengan nama umum Sunda Island pit viper. Banyak peneliti mencoba mengembangkan sebuah senyawa untuk menetralisir efek patologis dari gigitan ular. Sebuah molekul kecil alami serta sintesis varespladib banyak dikaji untuk perkembangannya dalam menghambat salah satu komponen utama venom, yaitu enzim PLA2. Penelitian ini bertujuan untuk menentukan efektivitas senyawa varespladib dalam menetralisir efek hemoragi yang diinjeksi oleh venom T. insularis pada mencit dan mengamati perubahan kondisi histopatologi kulit mencit setelah injeksi venom T. insularis serta perlakuan dengan varespladib. Pada penelitian ini dilakukan metode scoring pada sampel kulit dan histopatologi kulit mencit. Berdasarkan penelitian diperoleh hasil berupa penurunan tingkat hemoragi makroskopis secara signifikan ada pada kombinasi venom:varespladib sebesar 2:20 MHD dan penurunan tingkat hemoragi mikroskopis secara signifikan ada pada kombinasi venom:varespladib sebesar 2:10 dan 2:20 MHD. Terdapat perubahan rasio ketebalan lapisan dermis dan hipodermis secara signifikan melalui injeksi venom:varespladib. Adanya tren kenaikan jumlah sel inflamasi pada kulit mencit yang diinjeksi venom T. insularis serta tren penurunan jumlah sel inflamasi pada injeksi campuran venom:varespladib. Muncul kerusakan kolagen pada histopatologi kulit mencit yang diinjeksi venom T. insularis dengan tren yang naik dan turun bersamaan dengan injeksi campuran venom:varespladib.
Snake bites are a serious health problem in many countries, especially in tropical countries such as Southeast Asia, including Indonesia. According to data from the World Health Organization in 2024, snake bites are estimated to have caused approximately 81,000 to 138,000 deaths worldwide, with 400,000 cases resulting in permanent disability. One of the venomous snake species commonly found in Indonesia is Trimeresurus insularis, commonly known as the Sunda Island Pitviper. Many researchers have attempted to develop a compound to neutralize the pathological effects of snake bites. A small natural and synthetic molecule called varespladib has been extensively studied for its ability to inhibit one of the main components of venom, namely the PLA2 enzyme. This study aims to determine the effectiveness of the varespladib compound in neutralizing the hemorrhagic effects injected by T. insularis venom in mice and to observe changes in the histopathological condition of the mice's skin after injection of T. insularis venom and treatment with varespladib. In this study, a scoring method is used on skin samples and the histopathology of the mice's skin. Based on the research, the results are obtained a significant decrease in the level of macroscopic hemorrhage in the combination of venom:varespladib by 2:20 MHD and a significant decrease in the level of microscopic hemorrhage in the combination of venom:varespladib by 2:10 and 2:20 MHD. There is a significant change in the thickness ratio of the dermis and hypodermis layers through the injection of venom:varespladib. There is a trend of increasing the number of inflammatory cells in the skin of mice injected with T. insularis venom and a trend of decreasing the number of inflammatory cells in the injection of the venom:varespladib mixture. Collagen damage appeared in the histopathology of the skin of mice injected with T. insularis venom with an increasing and decreasing trend along with the injection of the venom:varespladib mixture.
