Identifikasi Penyebab dan Evaluasi Ketahanan Varietas Teh Assamica terhadap Penyakit Akar Putih

Penyakit akar putih merupakan salah satu penyakit penting pada tanaman teh (Camellia sinensis var. assamica) yang dapat menyebabkan kerugian cukup besar karena kematian tanaman. Penyakit tersebut disebabkan oleh jamur patogen tular tanah yang menginfeksi perakaran tanaman dan menyebabkan terganggunya penyerapan air serta unsur hara bagi tanaman. Penelitian ini bertujuan untuk mengetahui jamur penyebab penyakit akar putih pada tanaman teh di perkebunan PT. Pagilaran, Jawa Tengah, serta mengetahui tingkat ketahanan tiga klon teh assamica terhadap infeksi patogen tersebut. Metode yang dilakukan dalam penelitian ini adalah isolasi jamur dari akar tanaman bergejala, identifikasi molekuler menggunakan teknik PCR dengan pasangan primer ITS1 dan ITS4, serta uji postulat Koch untuk memastikan patogenisitas isolat jamur yang diperoleh. Persentase penyakit yang disebabkan oleh jamur patogen akar tersebut berkisar antara 2,48% hingga 32,83%. Uji postulat Koch membuktikan bahwa isolat jamur yang diperoleh dapat menimbulkan gejala penyakit yang sama, sehingga isolat jamur tersebut memenuhi kriteria sebagai patogen penyebab penyakit akar putih. Hasil analisis pengurutan sekuens menunjukkan bahwa isolat jamur yang ditemukan memiliki kemiripan tinggi dengan Xylaria apoda, yang memiliki tingkat kemiripan berdasar wilayah ITS sebesar 98–99?ngan data sekuens yang terdapat di GenBank. Evaluasi ketahanan terhadap tiga klon teh (PGL 10, PGL 15, dan TRI 2025) menunjukkan adanya perbedaan yang signifikan dari ketiga klon pada perlakuan inokulasi jamur. Klon PGL 10 menunjukkan intensitas penyakit yang terendah sebesar 63,33%, PGL 15 sebesar 66,67?n intensitas penyakit yang tertinggi pada klon TRI 2025 sebesar 73,33%. Berdasarkan tingkatan intensitas penyakit tersebut, seluruh klon dikategorikan dalam kategori ketahanan yang rentan.

White root disease is one of the important diseases in tea plants (Camellia sinensis var. assamica) which can cause significant losses due to plant death. The disease is caused by a soil-borne pathogenic fungus that infects plant roots and disrupts water as well as nutrient absorption for the plant. This study aims to identify the fungal pathogen that causes white root disease in tea plants at the PT. Pagilaran plantation, Central Java, and to evaluate the level of resistance of three assamica tea clones to this pathogenic infection. The methods used in this study were fungal isolation from the roots of symptomatic plants, molecular identification using PCR techniques with ITS1 and ITS4 primer pairs, and Koch's postulate test to confirm the pathogenicity of the fungal isolates. The percentage of disease caused by the root pathogen ranged from 2.48% to 32.83%. Koch's postulates confirmed that the fungal isolates can cause the same disease symptoms, thus the fungal isolate meets the criteria for a white root disease pathogen. Results of the sequence analysis showed that the fungal isolates had a high similarity to Xylaria apoda, which had a similarity level based on the ITS region about 98–99% compared with sequence data found in GenBank. Resistance evaluation of three tea clones (PGL 10, PGL 15, and TRI 2025) showed significant differences between clones in the fungal inoculation treatment. PGL 10 had the lowest disease intensity at 63.33%, followed by PGL 15 at 66.67%, and TRI 2025 at 73.33%. Based on disease intensity, all clones were classified as susceptible.
