Latar Belakang: Sistem imun bawaan berperan dalam mempertahankan tubuh terhadap infeksi dan menjaga homeostasis. Penurunan fungsi sistem imun dapat meningkatkan kerentanan terhadap penyakit, sehingga diperlukan agen imunostimulan yang efektif dan aman. Sediaan bahan alam diketahui memiliki potensi imunostimulan, namun mekanisme imunologis dan molekuler yang mendasarinya masih terbatas dikaji.
Tujuan: Penelitian ini bertujuan untuk mengetahui pengaruh pemberian sediaan bahan alam imunostimulan terhadap kadar nitric oxide (NO), aktivitas myeloperoxidase (MPO), serta ekspresi mRNA nitric oxide synthase 2 (NOS2) pada mencit BALB/c.
Metode: Penelitian ini merupakan penelitian eksperimental in vivo menggunakan mencit jantan BALB/c yang dibagi menjadi empat kelompok, tujuh ekor perkelompok, yaitu kelompok kontrol dan tiga kelompok perlakuan dengan dosis 592 mg/kgBB, 789 mg/kgBB, dan 1183 mg/kgBB yang diberikan secara oral selama 14 hari. Kadar NO dan aktivitas MPO dianalisis dari makrofag cairan peritoneal, sedangkan ekspresi mRNA NOS2 dianalisis pada jaringan hepar menggunakan metode rt-PCR.
Hasil: Hasil penelitian menunjukkan kadar nitric oxide (NO) meningkat secara signifikan pada dosis 789 mg/kgBB dan 1183 mg/kgBB dibandingkan kelompok kontrol (p<0 xss="removed">Kesimpulan: Sediaan bahan alam imunostimulan dapat meningkatkan respons imun bawaan melalui peningkatan kadar NO pada dosis 789 mg/kgBB, dan 1183 mg/kgBB, aktivitas MPO pada dosis 789 mg/kgBB, dan ekspresi mRNA NOS2 pada dosis 592 mg/kgBB, 789 mg/kgBB, dan 1183 mg/kgBB yang diberikan pada mencit BALB/c secara oral.

Background: The innate immune system plays a crucial role in defending the body against infections and maintaining homeostasis. A decline in immune function can increase susceptibility to disease; therefore, effective and safe immunostimulatory agents are needed. naturalformulations are known to have immunostimulatory potential; however, the underlying immunological and molecular mechanisms remain insufficiently explored.
Purpose: This study aimed to evaluate the effects of a polyherbal immunostimulant formulation on nitric oxide (NO) levels, myeloperoxidase (MPO) activity, and nitric oxide synthase 2 (NOS2) mRNA expression in BALB/c mice.
Methods: This was an in vivo experimental study using male BALB/c mice divided into four groups, seven mice were include in each group: a control group and three treatment groups receiving oral doses of 592 mg/kg body weight (BW), 789 mg/kg BW, and 1183 mg/kg BW for 14 days. NO levels and MPO activity were analyzed in peritoneal fluid macrophages, while NOS2 mRNA expression was analyzed in liver tissue using quantitative reverse transcription polymerase chain reaction
(rt-PCR).
Results: The results showed that nitric oxide (NO) levels increased significantly at doses of 789 mg/kg BW and 1183 mg/kg BW compared with the control group (p < 0 xss="removed">Conclusion: The naturals immunostimulant formulation enhances innate immune responses by increasing NO production at doses of 789 mg/kg BW and 1183 mg/kg BW, MPO activity at a dose of 789 mg/kg BW, and NOS2 mRNA expression at doses of 592 mg/kg BW, 789 mg/kg BW, and 1183 mg/kg BW following oral administration in BALB/c mice.
