ABSTRAK
Delayed wound healing in patients with diabetes mellitus (DM) is often caused by impaired circulation and an elevated risk of infection. Piper crocatum leaf extract, rich in synergistic flavonoids, holds promise for enhancing tissue regeneration, yet comprehensive studies on its molecular mechanisms and optimal formulation for improved bioavailability remain scarce. This study evaluates the therapeutic potential of ethanol extract of P. crocatum formulated as a SNEDDS gel in accelerating diabetic wound closure in BALB/c mice and characterizes flavonoid multitarget interactions via network pharmacology. The extract was obtained by ethanol maceration and subsequently incorporated into a SNEDDS gel using Tween 80 and propylene glycol at a 3:1 surfactant:cosurfactant ratio, yielding the optimal F3 formulation (Grade A) with a uniform particle size distribution. In vivo assessments included induction of hyperglycemia, measurement of wound closure rate, and collagen deposition via Sirius Red staining on days 7 and 14. Histometric analysis showed that treated groups exhibited significantly faster wound closure and increased collagen deposition (46.12 ± 4.44% on day 7; 60.36 ± 13.92% on day 14; p < 0>P. crocatum SNEDDS gel significantly accelerates diabetic wound healing through integrated multitarget mechanisms and enhanced collagen deposition, supporting its development as a flavonoid-based therapeutic for chronic wound management in DM.
