PENGARUH PENAMBAHAN ANTIBIOTIK GENTAMICIN, PENICILLIN DAN STREPTOMYCIN DALAM LARUTAN PENGENCER NaCl FISIOLOGIS TERHADAP KUALITAS SPERMATOZOA AYAM PELUNG YANG DISIMPAN PADA SUHU 5oC

2011 | Skripsi | S1 ILMU DAN INDUSTRI PETERNAKANPenelitian ini bertujuan untuk mengetahui pengaruh penambahan antibiotik Gentamicin, Penicillin, dan Streptomycin dalam larutan pengencer NaCl fisiologis terhadap pH, motilitas, viabilitas, dan abnormalitas spermatozoa dalam penyimpanan suhu 5oC. Sperma diambil setiap 4 hari sekali dari 5 ekor ayam pelung jantan berumur lebih dari satu tahun. Sperma yang diperoleh dibagi menjadi 4 perlakuan, yaitu kontrol (tanpa antibiotik), P1 (penambahan Gentamicin), P2 (penambahan Penicillin), dan P3 (penambahan Streptomycin). Setiap perlakuan disimpan pada suhu 5oC, kemudian diamati pada jam ke-0, 3, 6, 9, 24. Data dianalisis menggunakan Rancangan Split Subjek, dilanjutkan dengan uji DMRT. Hasil penelitian menunjukkan bahwa perlakuan penambahan antibiotik dalam pengencer memberikan pengaruh tidak nyata terhadap pH sperma, tetapi lama penyimpanan berpengaruh nyata (p?0,05) terhadap pH sperma. Perlakuan penambahan antibiotik dalam pengencer dan lama penyimpanan berpengaruh nyata (p?0,05) terhadap motilitas, viabilitas, dan abnormalitas spermatozoa ayam pelung. Rerata pH sperma perlakuan kontrol, P1, P2, P3 berurutan adalah 7,47±0,35; 7,54±0,36; 7,38±0,34; 7,25±0,35. Rerata motilitas spermatozoa perlakuan kontrol, P1, P2, P3 berurutan adalah 47,57±22,50%; 66,86±19,02%; 33,43±26,40%; 61,43±19,20%. Rerata viabilitas spermatozoa perlakuan kontrol, P1, P2, P3 berurutan adalah 50,96±24,90%; 62,44±21,45%; 37,84±26,46%; 54,77±21,37%. Rerata abnormalitas spermatozoa perlakuan kontrol, P1, P2, P3 berurutan adalah 49,04±24,61%; 44,67±24,07%; 53,99±23,82%; 44,39±18,63%. Dari hasil penelitian dapat disimpulkan bahwa penambahan Gentamicin dalam NaCl fisiologis lebih mampu mempertahankan motilitas dan viabilitas spermatozoa, penambahan Streptomycin dalam NaCl fisiologis lebih mampu menurunkan jumlah pertambahan spermatozoa abnormal, akan tetapi perlakuan penambahan antibiotik tidak memberikan pengaruh yang berbeda terhadap pH sperma. THE EFFECT OF ADDING GENTAMICIN, PENICILLIN AND STREPTOMYCIN ANTIBIOTICS IN PHYSIOLOGICAL NaCl DILUTERS ON DOMESTIC COCKS (PELUNG) SPERMATOZOA QUALITY STORAGED AT TEMPERATURE 5oC HELENA CHARISMA KRISTI 07/256628/PT/05384 ABSTRACT The objective of this research was to verify the effect of adding Gentamicin, Penicillin, and Streptomycin antibiotics in physiological NaCl diluters on the pH, motility, viability, and abnormality of spermatozoa storage at temperature 5oC. Semen was collected and pooled once time every 4 days from 5 cockerels aged more than one year old. Semen devided into four treatments, there were control (without antibiotic), P1 (adding the Gentamicin), P2 (adding the Penicillin), and P3 (adding the Streptomycin). All semen treatment storage at temperature 5oC, then evaluated after 0, 3, 6, 9, and 24 hours storage. Data were analyzed with Split Subject Design and Duncan’s Multiple Range Test (DMRT). The result showed that antibiotic adding treatment in diluters did not influenced to semen pH, but storage time of semen affected significantly different (p?0,05) to semen pH. Antibiotic adding treatment in diluters and storage time of semen affected significantly different (p?0,05) on motility, viability, and abnormality spermatozoa. pH semen in treatment control, P1, P2, and P3 were 7.47±0.35; 7.54±0.36; 7.38±0.34; and 7.25±0.35 respectively. Motility spermatozoa in treatment control, P1, P2, and P3 were 47.57±22.50%; 66.86±19.02%; 33.43±26.40%; and 61.43±19.20% respectively. Viability spermatozoa in treatment control, P1, P2, and P3 were 50.96±24.90%; 62.44±21.45%; 37.84±26.46%; and 54.77±21.37% respectively. Abnormality spermatozoa in treatment control, P1, P2, and P3 were 49.04±24.61%; 44.67±24.07%; 53.99±23.82%; and 44.39±18.63% respectively. It was concluded that Gentamicin adding in physiological NaCl more able to maintain the motility and viability spermatozoa, Streptomycin in physiological NaCl more able to reduce amount of adding abnormality spermatozoa, however antibiotic adding treatment not give the different to semen pH.
