Optimasi Produksi Kitosan dari Kitin Selongsong Maggot Menggunakan Sonikasi Melalui Response Surface Methodology

The sonochemical deacetylation of chitin isolated from maggot pupal exuviae was optimized using Response Surface Methodology (RSM). The objectives were to perform the deacetylation of chitin using sonication to accelerate the reaction time and optimize the deacetylation conditions by varying NaOH concentration (% b/v), temperature (°C), and sonication time (minutes) within an RSM-Central Composite Design (CCD) framework. The RSM-CCD method was employed to analyze the experimental data, and the degree of deacetylation (DD) was determined from Fourier Transform Infrared (FTIR) spectra using baseline correction.
The results indicated that the yield of chitin from maggot pupal exuviae was 25.21%, and the produced chitosan exhibited a DD range between 37.86% and 75.08%. Further, optimization through RSM-CCD revealed the optimal deacetylation conditions to be 60% NaOH concentration, 70°C temperature, and 120 minutes of sonication, achieving an optimum DD of 74.80%. These conditions demonstrate that the combination of these variables significantly enhances the degree of deacetylation.
