PENGARUH PENYIMPANAN SPERMA AYAM KAMPUNG YANG DIENCERKAN DENGAN LARUTAN PHOSPHATE BUFFER SALINE (PBS) PADA SUHU 4 SAMPAI 5° C SELAMA 24 JAM TERHADAP KUALITAS SPERMA DAN FERTILITAS TELUR

2003 | Skripsi | S1 PETERNAKANPenelitian ini bertujuan untuk mengetahui pengaruh penyimpanan sperma pada suhu 4 sampai 5° C selama 24 jam terhadap kualitas sperma dan fertilitas telur. Sperma dikoleksi dari 10 ekor ayam kampung jantan yang berumur 20 sampai 22 minggu dengan metode Burrow and Quin. Perlakuan pada penelitian ini meliputi sperma segar dan sperma simpan pada suhu 4-5° C selama 24 jam. Masingmasing perlakuan tersebut diencerkan dengan phosphate buffer saline (PBS) dengan perbandingan 1:2. Inseminasi buatan (IB) dengan dosis 100 juta sel sperma/ml diberikan kepada 10 ekor ayam petelur strain Lohman Brown untuk uji fertilitas. Koleksi telur dilakukan pada hari kedua setelah inseminasi selama satu minggu pada semua perlakuan. Variabel yang diamati meliputi kuantitas dan kualitas sperma segar yang terdiri dari volume sperma (ml), derajat keasaman (pH), konsentrasi (109/ml), motilitas spermatozoa (%) dan kualitas sperma simpan yang meliputi motilitas sperma simpan (%), derajat keasaman (pH) sperma simpan serta fertilitas telur sperma segar dan simpan (%). Data yang diperoleh dianalisis dengan analisis variansi menggunakan Rancangan Acak Lengkap Pola Searah. Hasil penelitian menunjukkan bahwa penyimpanan sperma pada suhu 4 sampai 5° C selama 24 jam berpengaruh sangat nyata (P<0,01) terhadap penurunan pH, motilitas dan fertilitas telur yang masing-masing adalah 6,6; 73,23% dan 65,45%. Dapat disimpulkan bahwa penyimpanan sperma pada suhu 4 sampai 5° C selama 24 jam akan menurunkan pH sperma, motilitas dan fertilitas.Kata kunci : Penyimpanan, Sperma, PBS, Fertilitas, Ayam kampung.
The effect of native cockerel sperms conservation on Phosphate Buffer Saline (PBS) diluted solution during 24 hours at 4 to 5 0 C temperature on sperm quality and egg fertility Dwi Fajaryadi 98/121353/PT/036X4 Abstract The objective of this study was to investigate the effect of native cockerel sperm conservation on PBS diluted at 4 to 5°C temperature during 24 hours on sperm quality and egg fertility. Sperms were collected from ten cocks, twenty-two weeks old of age. Fresh sperm and conserved was diluted with PBS, ratio of 1:2. The sperm were inseminated to ten hen layers of Lohman Brown strain with the dose of 100 million spermatozoa/ml for fertility test. The eggs were collected on second day after insemination for one weeks for all treatments. Variabels measured were quantity and quality of fresh sperms namely sperm volume (ml), degree of acidity (pH), concentration (109/ml), sperm motility (%) and conserved sperm quality which included conserved sperm motility (%), conserved sperm degree of acidity (pH) also egg fertility of fresh sperm and conserved sperm (%). Data were analyzed by varians analyze using Completely Randomized Design One-way Classification. The results indicated that conservation sperm at 4 , to 5°C temperature during 24 hours was significantly (P< 0.01) decreasing pH, motility and fertility of eggs, each of these were 6.6; 73.23 %; and 65.45 %. It was concluded that conservation of sperm at 4 to 5°C temperature during 24 hours would decrease of sperm pH, motility and fertility.
