Pengaruh Penambahan Selenium pada Medium Pengencer Komersial Terhadap Kualitas Semen Post-thawing Kambing Saanen

Prosesing semen beku dapat menyebabkan kerusakan spermatozoa akibat terbentuknya radikal bebas. Selenium sebagai antioksidan berpotensi menangkal efek buruk dari radikal bebas tersebut. Penelitian ini bertujuan untuk mengetahui pengaruh penambahan selenium terhadap kualitas semen post-thawing kambing Saanen. Penelitian dilaksanakan pada bulan Desember 2024 hingga Maret 2025 di Laboratorium Fisiologi dan Reproduksi Ternak, Fakultas Peternakan, Universitas Gadjah Mada. Pembuatan semen-selenium beku dilakukan di Balai Inseminasi Buatan Lembang, Jawa Barat. Semen dikoleksi dari pejantan unggul kambing Saanen dan dievaluasi berdasarkan standar kelayakan sebelum diproses pengenceran dan pembekuan. Semen kemudian disimpan dalam nitrogen cair pada suhu -196°C. Perlakuan yang diberikan berupa penambahan selenium pada medium pengencer komersial yang terbagi menjadi tiga level yaitu, 0 ppm (P0), 5 ppm (P1), dan 10 ppm (P2). Semen beku kemudian dicairkan pada suhu 37°C dalam water bath selama 30 detik sebelum dilakukan evaluasi kualitas post-thawing. Setiap perlakuan diuji dalam tujuh ulangan. Parameter yang diamati meliputi motilitas, viabilitas, abnormalitas, integritas membran plasma, dan integritas tudung akrosom. Data dianalisis menggunakan one-way analysis of variance (ANOVA) rancangan acak lengkap (RAL). Hasil penelitian menunjukkan bahwa penambahan selenium berpengaruh signifikan (p<0>0.05). Rata-rata motilitas (%) pada P0, P1, dan P2 yaitu, 55,00±2,89, 67,86±2,67, dan 68,57±2,44. Viabilitas (%) sebesar 77,78±3,85, 91,50±1,00, dan 91,10±1,97; sedangkan integritas membran plasma (%) sebesar 93,28±1,50, 95,78±1,60, dan 95,28±1,44. Integritas tudung akrosom (%) sebesar 94,86±2,13, 97,07±0,67, dan 95,14±1,07. Berdasarkan hasil penelitian, dapat disimpulkan bahwa penambahan selenium dapat meningkatkan kualitas semen post-thawing kambing Saanen.

The process of freezing semen can cause significant damage to spermatozoa due to the formation of free radicals. Selenium, a potent antioxidant, has the potential to mitigate these detrimental effects. This study aimed to evaluate the impact of selenium supplementation on post-thaw quality of Saanen buck semen. The research was conducted from December 2024 to March 2025 at the Laboratory of Animal Physiology and Reproduction, Faculty of Animal Science, Universitas Gadjah Mada. The production of selenium-supplemented frozen semen was conducted at the Lembang Artificial Insemination Center, West Java. Semen was collected from a superior Saanen buck and subjected to standard evaluation criteria before undergoing dilution and freezing. The semen were stored in liquid nitrogen at a temperature of -196°C. The experimental treatments involved the addition of selenium to the comercial diluent medium at three different concentrations: 0 ppm (P0), 5 ppm (P1), and 10 ppm (P2). Frozen semen was thawed at 37°C in a water bath for 30 seconds before post-thaw quality assessment. Each treatment was tested in seven replications. The assessed parameters included motility, viability, abnormality, plasma membrane integrity, and acrosome integrity. The data were statistically analyzed using the one-way analysis of variance (ANOVA) completely randomized design (CRD). The results showed that the addition of selenium had a significant effect (p<0>0.05). The mean value of motility (%) for P0, P1, and P2 were 55.00±2.89, 67.86±2.67, and 68.57±2.44 respectively. Viability (%) were 77.78±3.85, 91.50±1.00, and 91.10±1.97; while plasma membrane integrity (%) were 93.28±1.50, 95.78±1.60, and 95.28±1.44. Acrosome integrity values (%) were 94.86±2.13, 97.07±0.67, and 95.14±1.07. Based on the result of the study, it can be concluded that the addition of selenium enhances post-thaw semen quality in Saanen bucks.
