Analisis Keragaman Genetik Ikan Tenggiri (Scomberomorus sp.) Asal Cilacap, Rembang, Fakfak, dan Manggarai Barat Berdasarkan Sekuen Gen Cytochrome B

Mackerels or Scomberomorus sp. is a pelagic fish species of superior fishery commodities that has high economic value in Indonesia. Market demand and public purchasing power for mackerel and its processed products which tend to increase every year can trigger uncontrolled fishing which results in population decline so that genetic diversity analysis is needed as a basis for determining management policies in observing mackerel population stability. This study aims to identify kinship and species variation in mackerel from Cilacap, Rembang, Fakfak, and West Manggarai based on cytochrome b (cyt b) encoding genes in mitochondrial DNA. Samples were amplified by PCR method using primers forward cytBF and reverse cytBR resulting in PCR products of 1.141 bp which were then sequenced. Sequencing results were analyzed by comparing samples with the database of comparator species from GenBank. Sequencing results in the form of 1141 nucleotide sites encoding 380 amino acids, Based on the results of the analysis, it is known that the mackerel samples from Cilacap (C1, C2, C3) were identified as Scomberomorus guttatus (PP437201. 1) with average genetic distance of 1,69%; samples from Rembang (R1, R2, R3), Papagarang Island (P1A, P2A, P3A), and two samples from Labuan Bajo (B2 and B3) were identified as Scomberomorus commerson (PP43720) with average genetic distance of 0,18%-1,06%; samples from Fakfak (P1B, P2B, P3B) were identified as Scomberomorus semifasciatus strain GREY-SsPD211135 (NC021392.1) with average genetic distance of 0,26%; and one sample from Labuan Bajo (B1) was identified as Acanthocybium solandri (OP035261.1) with average genetic distance of 0,26%.
