Pengaruh ekstrak etanolik Sargassum sp. terhadap kadar Matrix metalloproteinase-8 pada periodontitis diabetika (kajian in vivo pada Rattus norvegicus)

Periodontitis adalah inflamasi pada jaringan pendukung gigi, yang dapat diperparah dengan adanya keterlibatan penyakit diabetes melitus (DM). Matrix metalloproteinase-8 (MMP-8) adalah salah satu marker potensial yang dapat menandai kerusakan kolagen dan matriks ekstraseluler pada jaringan periodontal terkait periodontitis diabetika. Ekstrak etanolik Sargassum sp. dengan berbagai kandungan aktif didalamnya dapat menekan respon inflamasi yang terjadi. Tujuan penelitian ini adalah untuk mengetahui pengaruh ekstrak Sargassum sp. terhadap kadar MMP-8 pada cairan sulkus gingiva periodontitis diabetika Rattus norvegicus.
Subjek penelitian terdiri dari 24
ekor tikus Rattus norvegicus yang
terbagi dalam 4 kelompok, yaitu tikus normal, kelompok periodontitis diabetika yang
diberikan NaCMC (K-), kelompok periodontitis diabetika yang
diberikan ekstrak Sargassum sp. 70% dosis 300 mg/kg BB dan kelompok
periodontitis diabetika yang diberikan obat doxycycline (K+).
Sebelum perlakuan, terlebih dahulu tikus diinduksi DM dengan penyuntikan STZ secara
intraperitoneal. Setelah 3 hari, induksi periodontitis dilakukan dengan ligasi
menggunakan benang silk 4.0 dan injeksi LPS Aa. Setelah 15 hari perlakuan,
kadar MMP-8 cairan sulkus gingiva masing-masing subjek diambil dan diuji
menggunakan ELISA.
Hasil uji analisis statistik one-way
ANOVA menunjukkan bahwa terdapat pengaruh pemberian ekstrak Sargassum sp.
pada kadar MMP-8 cairan sulkus gingiva (p<0>post-hoc
LSD juga menunjukkan bahwa pemberian ekstrak Sargassum sp. memberikan
pengaruh yang setara dengan pemberian doxycycline (p>0,05) sebagai
kontrol positif. Kesimpulan penelitian ini adalah bahwa ekstrak etanolik
Sargassum sp. 70?rpengaruh menurunkan kadar MMP-8 pada model
periodontitis diabetika Rattus norvegicus.
Periodontitis is inflammation of
supporting periodontal tissue which can exacerbated by involvement of diabetes
mellitus disease (DM). Matrix metalloproteinase-8 (MMP-8) is the one of potential
marker related diabetic periodontitis. Ethanolic extract of Sargassum sp.
with various active inggredients can
suppress the inflammatory response. The purpose of this study was to determine
the effect of Sargassum sp. extract on MMP-8 levels in gingival
crevicular fluid of diabetic periodontitis (Rattus novergicus).
Subject of this study consisted of 24 Wistar rats divided into 4 groups, namely normal rats, diabetic periodontitis group given by NaCMC (K-), diabetic periodontitis group given ethanol extract of Sargassum sp. 70% and diabetic periodontitis group given by doxycycline (K+). Before treatment, subjects were first induced with DM by intraperitoneal injection of STZ. After 3 days, periodontitis induction was carried out by ligation using silk thread 4.0 and injection of LPS Aa. After 15 days of treatment, MMP-8 levels of the gingival crevicular fluid of each subject were taken and tested using ELISA.
The results of one way ANOVA statistical analysis test showed that there was an effect of giving Sargassum sp. extract on the MMP-8 levels of gingival crevicular fluid (p<0>Sargassum sp. extract had an effect equivalent to giving doxycycline as positive control (p>0,05). The conclusion of this study is that extract of Sargassum sp. 70% is effective in reducing MMP-8 levels in the diabetic periodontitis Rattus norvegicus.
