Pengaruh Pemberian Sekretom dan Eksosom pada Perancah Gelatin Terhadap Jumlah Sel Osteoblas dan Osteosit Sebagai Perawatan Pre-Prostodontik

Tooth extraction leads to a decrease in alveolar bone volume, impacting the retention and stability of dentures. Secretomes and exosomes derived from dental pulp stem cells are believed to have regenerative properties for bone tissue. This study investigates the effect of adding these materials to gelatin scaffolds on the formation of osteoblasts and osteocytes, which are indicators of bone regeneration, as a pre-prosthodontic treatment.
An experimental laboratory study was conducted using 3-month-old male Wistar rats weighing between 200-250 mg. A defect measuring 2.5x2.5x3 mm was created in the right femoral condyle. The study included four groups: control (defect only), P1 (gelatin scaffold), P2 (gelatin scaffold with secretome), and P3 (gelatin scaffold with exosomes). The number of osteoblasts and osteocytes was observed on days 14 and 28 using Hematoxylin Eosin staining, analyzed under a light microscope at 400x magnification, and quantified with ImageJ software using two-way ANOVA.
Results indicated that secretomes and exosomes significantly increased the number of osteoblasts and osteocytes compared to the control group and P1 at both 14 and 28 days post-application. While the combination of secretome and gelatin scaffold (P2) showed a greater effect than exosomes with gelatin scaffold (P3), this difference was not statistically significant. The conclusion drawn is that secretomes and exosomes can enhance osteoblast and osteocyte numbers, supporting their use in pre-prosthodontic treatments.
