Pengaruh Gel Ekstrak Etanolik Gama Melon Parfum (Cucumis melo L. cv. ‘GMP’) terhadap Jumlah Neutrofil pada Model Periodontitis (Kajian In Vivo pada Tikus Wistar)

Periodontitis is a chronic inflammatory disease that causes damage to periodontal tissues through a complex interaction between pathogenic bacteria, host responses, and accompanying risk factors. The administration of anti-inflammatory agents as an adjuvant to scaling and root planing (SRP) aims to control the inflammatory phase and prevent it from becoming prolonged. Gama Melon Parfum (GMP) is a melon cultivar developed by the Faculty of Biology UGM which contains various anti-inflammatory phytochemical components. This study aimed to determine the effect of ethanol extract gel from Gama Melon Parfum (Cucumis melo L. cv. ‘GMP’) in reducing the number of neutrophil in the Wistar rat periodontitis model.
The study was conducted by inducing periodontitis using the ligature method for 7 days in 24 Wistar rats. There were three research groups: a 10% GMP ethanol extract gel treatment group, a 5% GMP ethanol extract gel, and control group using base gel. The test materials were applied, and samples were collected on days 1, 3, 5, and 7 after application. Histological preparations were stained with hematoxylin-eosin, and the neutrophil count was calculated. The data were analyzed using the Shapiro-Wilk normality test and Levene’s test for homogeneity.
The average neutrophil count in all three groups showed a consecutive decline over the observation period. There was a significant difference in the average neutrophil count between the 10%, 5% ethanolic GMP extract gel groups, and the control group (p<0>0.05). The reduction in neutrophil count is thought to be due to the anti-inflammatory activity of flavonoids, cucurbitacin B, ascorbic acid, saponins, and terpenoids contained in GMP fruit. The conclusion is that GMP ethanol extract gel reduces neutrophil counts in the Wistar rat periodontitis model.
