PENAMBAHAN EKSTRAK HIPOFISIS KAMBING DENGAN DOSIS YANG BERBEDA PADA MEDIUM UNTUK MENINGKATKAN KEMAMPUAN MATURASI OOSIT KAMBING IN VITRO

Ovarium diambil dari Rumah Potong Hewan dan dibawa ke laboratorium dalam larutan NaCl fisiologis yang ditambah antibiotik pada suhu 31-34°C. Oosit diaspirasi menggunakan syringe 3 ml disposable yang telah diisi 1ml Dulbecco’s Phosphate Buffered Saline (DPBS). Pencucian oosit dilakukan di bawah mikroskop stereo. Oosit ditempatkan pada drop medium dan ditambahkan dengan ekstrak hipofisis dan diinkubasikan pada inkubator dengan suhu 39°C, kelembaban 95% dan kadar CO2 5% selama 24 jam. Oosit dibagi dalam tiga kelompok, yaitu dosis ekstrak hipofisis 0, 10, dan 20%. Data dianalisis menggunakan oneway ANOVA. Hasil penelitian menunjukkan terdapat perbedaan nyata pada persentase oosit mature dari ketiga kelompok perlakuan. Penambahan ekstrak hipofisis dengan dosis 20% menghasilkan prosentase maturasi terbesar (78,18±22,61%), diikuti dosis 10 dan 0% (65,15±23,5 dan 54,54±23,68%). Berdasarkan hasil penelitian dapat disimpulkan bahwa penambahan ekstrak hipofisis pada medium maturasi dapat meningkatkan kemampuan maturasi pada oosit kambing. ADDITION OF GOAT’S HYPOPHYSIS EXTRACT WITH DIFFERENT DOSES INTO MEDIUM TO IMPROVE IN VITRO MATURATION ABILITY OF GOAT’S OOCYTE Evita Yustian Putri 08/272757/PT/05598 ABSTRACT This study aimed to determine the effect of goat’s hypophysis extract addition into medium on the ability of goat oocytes to mature in vitro. Ovaries were collected from slaughterhouse and transported to the laboratory in physiological saline solution plus antibiotics at temperatures 31-34 ° C. Oocytes were aspirated using a 3 ml disposable syringe filled with 1ml Dulbecco's Phosphate Buffered Saline (DPBS). Oocyte were washed under a stereo microscope. Oocytes were placed in medium supplemented by pituitary extract and incubated in temperature of 39° C, 95% humidity and 5% CO2 for 24 hours. Oocytes were divided into three groups, i.e. the dose of hypophysis extract 0, 10, and 20%. Data were analyzed using oneway ANOVA. The results showed significant differences in the percentage of mature oocytes from the three treatment groups. The largest maturation percentage was shown when 20% hypophysis extract was added (78.18 ± 22.61%), followed by 10 and 0% (23.5 ± 65.15 and 54.54 ± 23.68%). Based on the results of this study, it could be concluded that the addition of hypophysis extract on maturation medium can improve the maturation ability of oocyte in goats.
