Sampel rennet disimpan dingin (8°C) dan beku (-20°C) selama 0, 15, dan 30 hari. Pengujian dilakukan terhadap waktu koagulasi susu, rendemen (curd), nilai pH, dan aktivitas proteolitik. Data yang diperoleh dianalisis dengan menggunakan variansi rancangan acak lengkap pola faktorial dilanjutkan Duncan’s New Multiple Range Test (DMRT). Hasil analisis menunjukkan bahwa penyimpanan rennet dingin dan beku hingga 30 hari berpengaruh nyata (P?0,05) terhadap waktu koagulasi susu, rendemen, nilai pH, dan aktivitas proteolitik dengan rerata total penyimpanan dingin dan beku waktu koagulasi 0 hari 201,33 detik, 15 hari 234,83 detik, 30 hari 278,17 detik. Rerata total rendemen 0 hari 11,07%, 15 hari 8,93%, 30 hari 7,47%. Rerata total pH 0 hari 5,6, 15 hari 5,88, 30 hari 5,94. Rerata total aktivitas proteolitik 0 hari 25,870 µg/ml/menit, 15 hari 24,798 µg/ml/menit, 30 hari 24,439 µg/ml/menit. Tidak ada perbedaan yang nyata antara penyimpanan dingin dan beku hingga 30 hari penyimpanan pada waktu koagulasi susu, rendemen (curd), dan nilai pH. Penyimpanan dingin dan beku berbeda nyata pada aktivitas proteolitik. Hal ini menunjukkan penyimpanan beku memiliki kemampuan lebih baik dalam mempertahankan aktivitas rennet lambung kelinci walaupun tidak mempertahankan waktu koagulasi, rendemen curd, dan pH yang lebih baik dibandingkan penyimpanan dingin. Penyimpanan dingin dan beku hingga 30 hari dapat menurunkan waktu koagulasi susu, rendemen curd, pH, dan aktivitas proteolitik rennet lambung kelinci. THE INFLUENCE OF CHILL AND FROZEN STORAGE ON PROTEOLYTIC ACTIVITY OF RENNET FROM RABBIT STOMACH By Mufti Tri Matra 05/186777/PT/04992 ABSTRACT This experiment was conducted to find out the influence of chill and rozen storage on proteolytic activity of rennet from rabbit stomach. Rennet samples were stored at chill (8°C) and frozen (-20°C) temperature or 0, 15 and 30 days. The parameters observed were milk coagulation ime, curd yield, pH, and proteolytic activity. The data was analyzed by using factorial completely randomized design continued with Duncan’s New Multiple Range Test (DMRT). The results of analysis showed that chill and frozen storage of rennet up to 30 days influenced (P?0,05) on milk coagulation time, curd yield, pH and proteolytic activity with total average of chill and frozen storage of milk coagulation time of 0 day was 201.33 seconds, 15 days was 234.83 seconds, 30 days was 278.17 seconds. The total average of curd yield of 0 day was 11.07%, 15 days was 8.93%, 30 days was 7.47%. The total averages of pH of 0 day was 5.6, 15 days was 5.88, 30 days was 5.94. The total average of proteolytic activity of 0 day was 25.870 µg/ml/minute, 15 days was 24.798 µg/ml/minute, 30 days was 24.439 µg/ml/minute. There was no significant difference between chill and frozen storage till 30 days on milk coagulation ime, curd yield and pH. Chill and frozen storage till 30 days was significantly difference on proteolytic activity. It indicated that frozen storage maintained proteolytic activity of rabbit stomach rennet better than chill storage although frozen storage was not better than chill storage on maintaining milk coagulation time, curd yield and pH. Chill and frozen storage up to 30 days degraded milk coagulation time, curd yield, pH and proteolytic activity of rennet from rabbit stomach.
