Background: Hypoxia is a condition of reduced thus lack of cellular oxygen supply. Repeated hypoxic conditions trigger excessive oxidative stress and inflammation, followed by activation of the homeostatic response through enzymatic and non-enzymatic antioxidants. Ficus carica is a fruit rich in antioxidants, thus may be predicted effectively suppress the oxidative stress and inflammation caused by induced chronic intermittent hypoxia (CIH). This study investigates the similar phenomena in the lung tissue.
Aim: To investigate the potential benefits of administering puree Ficus carica (PFC) amongst Sprague Dawley (SD) rats with CIH induction and its effect on the levels of Malondialdehyde (MDA), Superoxide Dismutase (SOD), SOD/MDA ratio, Platelet Lymphocyte Ratio (PLR), and Tumor Necrosis Factor-? (TNF-?).
Methods: This is an experimental design with a pre-posttest control and post-control. Thirty SD rats divided into six groups: neutral control (K), positive control given vitamin E (HRE), negative control (HR), and three PFC intervention groups with different doses 1.25, 2.5, 5.0 mL/200gBw/day respectively (HRF-1.25, HRF-2.50, and HRF-5.0). Except neutral control group, rats were on CIH with a cycle of four hours of hypoxia/day and 20 hours of reoxygenation for 30 days. ELISA method was used to measure MDA, SOD, and TNF-? levels in lung homogenate, while a hematology analyzer was used to measure blood PLR levels.
Results: The study found significant differences in lung homogenate levels between the HRF-5.0 and negative control groups. The MDA levels were lower (0.39±0.19 ng/mL) and SOD levels were higher (3.02±1.04 ng/mL) in the HRF-5.0 group compared to HR. The SOD/MDA ratio was higher (7.91±1.11 ng/mL) and TNF-? levels were lower (25.37±0.561 ng/mL) in the HRF-5.0 group compared to HR. Lower blood PLR levels (50.01±17.31 ng/mL) was found in HRF-5.0 compared to HR.
Conclusion: This study demonstrated the beneficial effect of PFC to suppress the level of pulmonary oxidative stress and inflammation induced by CIH in SD rats.
