STUDI MAGNETIC FLUORESCENT SILICA NANOPARTICLES (MFSNP) BERBASIS SILIKA GEOTHERMAL SEBAGAI BIOSENSOR DETEKSI BAKTERI S. AUREUS

Material magnetic fluorescent silica
nanoparticles (MFSNP) telah berhasil dibentuk dari 4 prekursor utama
yaitu silika oksida, besi oksida, agen fluoresens (rhodamin B) dan CTAB.
Keempat prekursor tersebut didesain menjadi 3 faktor (faktor Fe/Si, rhodamin B
dan CTAB) untuk menentukan 3 respon (kekuatan magnetis, intensitas fluoresens dan
surface area). Material MFSNP memiliki sifat bifungsional yaitu sifat
magnetis dan sifat fluoresens. Kedua sifat yang dimiliki oleh MFSNP dapat di
aplikasikan sebagai biosensor. Salah satu kegunaan dari biosensor MFSNP adalah
untuk deteksi baktri S. Aureus. Bakteri S. Areus merupakan
bakteri yang bersifat patogen yang hidup secara alami pada kulit mukosa
manusia. Berdasarkan dari hasil karakteristik pada pengujian XRD, material
MFSNP memiliki fase amorf pada silika oksida dan fase kristalin pada besi
oksida. Hasil pengujian STEM membuktikan
bahwa silika yang berasal dari geothermal telah berhasil melapisi besi
oksida membentuk nanokomposit. Hasil optimasi dari desain komposisi menggunakan
RSM menyatakan bahwa penggunaan faktor Fe/Si, rhodamin B dan CTAB secara
berurutan yang terbaik adalah 0,5 g/g, 5 mg/g dan 3%. Hasil percobaan dari
optimasi RSM diketahui bahwa MFSNP memiliki kekuatan magnetis sebesar 16,7 emu/g;
intensitas fluoresens sebesar 566,20 a.u.; dan surface area sebesar 139,45
m2/g. MFSNP optimum dari RSM dimodifikasi permukaannya menjadi MFSNP
termodifikasi vankomisin (MFSNP-Van) melalui reaksi undesililasi. MFSNP-Van
diaplikasikan untuk deteksi bakteri S. Aureus diketahui intensitas
sebelum dan setelahnya yaitu 76,52 a.u dan 5,45 a.u. dengan %Iloss
sebesar 92,88 ?n dilakukan pengujian
pembanding menggunakan bakteri MRSA diperoleh %Iloss sebesar 78,01
%.
Magnetic fluorescent silica
nanoparticles (MFSNP) have been successfully formed from 4 main precursors,
namely silica oxide, iron oxide, fluorescent agent (rhodamine B), and CTAB.
These four precursors were designed into 3 factors (Fe/Si factor, rhodamine B,
and CTAB) to determine 3 responses (magnetic strength, fluorescence intensity,
and surface area). MFSNP material has bifunctional properties, namely magnetic
and fluorescent properties. Both properties possessed by MFSNP can be applied
as a biosensor. One use of MFSNP biosensor is for the detection of S. Aureus
bacteria. S. Aureus bacteria are pathogenic bacteria that naturally live on
human mucosal skin. Based on the characteristic results of XRD testing, MFSNP
material has an amorphous phase in silica oxide and a crystalline phase in iron
oxide. STEM testing results prove that silica from geothermal sources has
successfully coated iron oxide to form a nanocomposite. The optimization
results of composition design using RSM state that the best use of Fe/Si,
rhodamine B, and CTAB factors sequentially is 0.5 g/g, 5 mg/g, and 3%. The
experimental results from RSM optimization show that MFSNP has a magnetic
strength of 16.7 emu/g; fluorescence intensity of 566.20 a.u.; and surface area
of 139.45 m2/g. The optimum MFSNP from RSM is surface-modified into
vancomycin-modified MFSNP (MFSNP-Van) through undesilylation reaction.
MFSNP-Van is applied for the detection of S. Aureus bacteria, and the intensity
before and after detection is 76.52 a.u. and 5.45 a.u., with a %Iloss of
92.88%. Comparative testing using MRSA bacteria resulted in a %Iloss of 78.01%.
