Pengambilan Selulosa Dari Tongkol Jagung dengan Deep Eutectic Solvent (DES) Untuk Meningkatkan Sifat Mekanis Bioplastik

Selulosa merupakan polisakarida yang ditemukan dalam
dinding sel tumbuhan dan merupakan komponen utama selain hemiselulosa dan
lignin dalam struktur biomassa seperti tongkol jagung. Selulosa memiliki
potensi untuk dimanfaatkan sebagai bahan pembuatan produk seperti pengembangan
bioplastik. Dengan karakteristik serat yang kuat, penambahan selulosa dapat
memperkuat matriks bioplastik, meningkatkan kekuatan tarik, kekakuan dan
ketahanan terhadap tekanan maupun panas. Selulosa dapat diambil dari tongkol jagung melalui pemutusan ikatan lignin menggunakan
metode ekstraksi padat-cair dengan Deep Eutectic Solvent (DES). DES
dibuat dengan mencampurkan choline chloride (ChCl) sebagai Hydrogen
Bond Acceptor (HBA) dan gliserol, urea, asam oksalat dihidrat serta asam
laktat sebagai variasi Hydrogen Bond Donor (HBD). HBA dan HBD dicampur
pada suhu 80°C dengan rasio molar tertentu disertai pengadukan 200 rpm selama 2
jam. Pengambilan selulosa menggunakan perbandingan
sampel:pelarut 1:20 (b/b). Variabel bebas yang dikaji meliputi variasi HBD DES,
rasio molar HBA:HBD (1:1, 1:2, 1:5, 1:8), suhu (90, 110, 130°C) dan waktu reaksi (2,
4, 6 jam). Kadar (%b/b) selulosa, hemiselulosa dan lignin dianalisis
menggunakan metode Chesson. Yield selulosa dioptimasi menggunakan Responce Surface Method (RSM)
untuk mendapatkan model persamaan yang sesuai dalam menggambarkan pengaruh
variabel bebas terhadap respon yield selulosa. Karakterisasi menggunakan
FTIR, XRD dan SEM dilakukan untuk mengetahui pengaruh DES terhadap intensitas,
kristalinitas dan morfologi selulosa. Untuk mendapatkan selulosa
berukuran kecil dan homogen digunakan metode
pengendapan ulang dan ultrasonikasi 600 Watt, 40 kHz selama 15 menit. Selulosa ditambahkan pada pembuatan bioplastik dari Poly
Lactic Acid (PLA) dan agen plasticizer gliserol menggunakan hydraulic hotpress pada tekanan 50
kg/mm2 dan suhu 180°C selama 10 menit.
Pengaruh penambahan selulosa terhadap sifat mekanis bioplastik diuji dengan mengukur elongasi menggunakan
UTM dan degradasi massa terhadap panas pada suhu 25-600°C menggunakan TGA.
Hasil penelitian diperoleh yield selulosa tertinggi 84,79% (b/b) dari DES ChCl:gliserol 1:5 pada suhu 110°C dan waktu reaksi 4 jam dengan ukuran partikel 66,7 nm. Optimasi yield selulosa dengan RSM menunjukkan model kuadratik merupakan model yang sesuai untuk menjelaskan pengaruh variabel bebas terhadap respon dengan p-value 0,0022 (signifikan) dan lack of fit 0,3653. Hasil karakterisasi FTIR dan XRD menunjukkan bahwa DES dapat mengurangi intensitas lignin dan meningkatkan intensitas serta kristalinitas selulosa. Hasil ini didukung oleh analisis morfologi permukaan dengan SEM yang menunjukkan serat selulosa terlihat lebih jelas setelah lignin berkurang. Hasil pengukuran elongasi dengan UTM dan degradasi massa dengan TGA membuktikan penambahan selulosa dapat meningkatkan kekakuan, kekerasan dan ketahanan panas bioplastik.
Cellulose is a polysaccharide found in the cell walls
of plants and is a major component, along with hemicellulose and lignin, in the
biomass structure of materials such as corn cobs. Cellulose has the potential
to be utilized as a material for various products, including its application in
the development of bioplastics. With its strong fiber characteristics, the
addition of cellulose can reinforce the bioplastic matrix, enhancing tensile
strength, stiffness, and resistance to pressure and heat. Cellulose can be
extracted from corn cobs by breaking the lignin bonds using the solid-liquid
extraction method with Deep Eutectic Solvent (DES). DES is made by mixing
choline chloride (ChCl) as Hydrogen Bond Acceptor (HBA) and glycerol, urea,
oxalic acid dihydrate and lactic acid as variations of Hydrogen Bond Donor
(HBD). HBA and HBD are mixed at 80°C with a specific molar ratio and stirred at
200 rpm for 2 hours. Cellulose extraction uses a sample:solvent ratio of 1:20
(w/w). Variables studied include variations in DES HBD, HBA:HBD molar ratio
(1:1, 1:2, 1:5, 1:8), temperature (90, 110, 130°C), and reaction time (2, 4, 6
hours). The content (%w/w) of cellulose, hemicellulose, and lignin is analyzed
using the Chesson method. Cellulose yield is optimized by Response Surface
Methodology (RSM) to obtain a suitable equation model describing the influence
of independent variables on the cellulose yield response. Characterisation by
FTIR, XRD, and SEM was performed to analyse the effect of DES on the intensity,
crystallinity, and morphology of the cellulose. To obtain small and homogenous
cellulose, the reprecipitation and ultrasonication method with 600 Watt and 40
kHz for 15 minutes is conducted. Cellulose is added in the production of
bioplastics from Poly Lactic Acid (PLA) and glycerol as plasticizer using
hydraulic hot press at pressure of 50 kg/mm2 and temperature of 180°C
for 10 minutes. The effect of adding cellulose on the mechanical properties of
bioplastics is tested by measuring elongation using UTM and mass degradation
due to heat at temperatures of 25-600°C using TGA.
The research results obtained the highest cellulose yield of 84.79% (w/w) from DES ChCl:glycerol 1:5 at 110°C and 4 hours of reaction time with particle size of 66.7 nm. Optimization of cellulose yield with RSM shows that the quadratic model is appropriate to explain the influence of independent variables on the response with p-value of 0.0022 (significant) and lack of fit 0.3653. FTIR and XRD characterization results show that DES can reduce lignin intensity and increase cellulose intensity and crystallinity. This is supported by surface morphology analysis with SEM, which shows cellulose fibers becoming clearer after lignin reduction. Measurement results of elongation with UTM and mass degradation with TGA prove that the addition of cellulose can increase the rigidity, hardness, and heat resistance of bioplastics.
