Breast cancer is the most frequently diagnosed cancer and is the leading cause of cancer-related deaths among women worldwide. Current chemotherapeutic agents have many limitations, including resistance, toxic effects, and weak efficacy. Nyamplung leaves (Calophyllum inophyllum) are one of six plants that have strong potential as anticancer agents from the screening results RISTOJA / Riset Tanaman Obat dan Jamu (National Research on Medicinal Plants and Herbs). Nyamplung leaf dichloromethane extract (NLDE) has cytotoxic activity against T47D breast cancer cells, WiDr colon cancer cells, and HeLa cervical cancer cells with the lowest IC50 being T47D cells. This study aims to explore the anticancer activity of the active fraction of NLDE on the T47D breast cancer cell line.
Nyamplung leaves were macerated with dichloromethane solvent then fractionated by decantation to separate n-hexane soluble and n-hexane insoluble compounds. All fractions were tested for cytotoxic activity by MTT Assay, and the most active fraction was further tested for advanced T47D cell apoptosis using flow cytometry with PI and Annexin V staining sequentially. Identification of compound groups in the active fraction was carried out by observing the TLC profile with spot appearance and analysis using LC-HRMS.
The NLDE fraction that has the most potent cytotoxic activity is the n-hexane insoluble fraction (HIF) with an IC50 of 23.41µg/mL against the T47D breast cancer cell line. HIF dose of 25.0 µg/mL increased the percentage of T47D cell apoptosis by 2.3%. Based on phytochemical profile analysis, HIF contains terpenoid, steroid and ortho dihydroxy flavonoid compounds. Overall, the HIF from NLDE has the potential to further explore other mechanisms of proliferation inhibition such as cell cycle inhibition, cell senescence or aging, and autophagy.
