Inceptisol is a type of soil that has developed quite a lot in various regions in Indonesia. Despite its very wide distribution in Indonesia, this land has several limitations, including low physical, chemical and biological soil fertility, and is exacerbated by drought conditions. The existence of drought conditions on land allows for a response to the cultivation of crops including Madagascar periwinkle, which is related to the provision of secondary metabolites. The aim of this research was to determine the level of soil fertility in the research area, the effect of the applications and doses of rice husk biochar (RHB), ZA fertilizer, and effective water stress Inceptisol in Karangwangkal, Banyumas, Central Java. The study was carried out experimentally using a split plot design with three treatment factors, each of which consisted of levels, namely 0, 15 ton.ha-1, and 30 ton.ha-1 for RHB, 0 kg.ha-1 120 kg.ha-1 and 240 kg.ha-1 for ZA fertilizer, and 0, 2, and 4 days watering interval for water stress treatment. The results showed that the application of ZA and RHB at doses of 30 ton.ha-1 and 240 kg.ha-1 respectively could increase, total nitrogen, pH, CEC, and soil organic matters. Apart from that, the best treatment for agronomic parameters, in form of plant height, number of leaves, total leaf area, fresh weight of roots and shoots, and dry weight of roots and shoots, were obtained in the applications of 30 ton.ha-1 of RHB, ZA fertilizer of 240 kg.ha-1, and watered every day. The secondary metabolite parameters vinblastine and proline showed different results. The best treatment for vinblastine metabolite parameters was obtained in 30 ton.ha-1 of RHB, 120 kg.ha-1 ZA fertilizer, and 4 days of watering interval with a level of 2104.52 µg/g. Meanwhile, proline contents were best in the application of 30 ton.ha-f of RHB, 240 kg.ha-1 ZA fertilizer, and 4 days of watering interval with levels of 25.20 µmol/g. It can be concluded that the production of secondary metabolites increases with the increasing water stress, the application of ZA fertilizer and RHB. The last two treatments could increase total nitrogen, pH, CEC, and soil organic matters, while daily watering will optimize the growth of periwinkle plants.
