Kemampuan Pseudomonas aeruginosa dalam Biosorpsi Tembaga dengan Penambahan Asam Amino L-Arginin

Salah satu pendekatan dalam mengatasi pencemaran adalah dengan memanfaatkan mikroorganisme dalam bioremediasi logam berat di lingkungan. Pseudomonas aeruginosa diketahui menunjukkan kemampuannya dalam mengurangi konsentrasi tembaga dengan menggunakan biofilm. Dalam penelitian ini, P. aeruginosa digunakan untuk menurunkan konsentrasi tembaga dengan membiakkan bakteri dalam medium cair Luria-Bertani (LB), ditambah dengan L-arginin pada konsentrasi 80 mM dan CuSO4 pada 750 ppm. Hasil penelitian menunjukkan bahwa medium dengan penambahan L-arginin meningkatkan pembentukkan biofilm sebesar 52,41% dibandingkan dengan perlakuan tanpa penambahan L-arginin. Analisis tembaga juga menunjukkan bahwa P. aeruginosa mampu menurunkan konsentrasi tembaga sebesar 145,54 ppm (20%). Ketika biomassa sel diaplikasikan untuk biosorpsi tembaga dalam larutan CuSO4 750 ppm (tanpa penambahan medium lain), ditemukkan bahwa konsentrasi tembaga dalam larutan berkurang sebesar 155,18 ppm (21%), dan 164,83 ppm (23%) setelah inkubasi masing-masing selama 14 jam dan 48 jam.

Copper is a heavy metal used in many industries and mining activities which has a potential to pollute the environment and becomes an imminent threats to health problem. One of the approaches in overcoming pollution is by employing microorganisms in bioremediating heavy metal excess amount in the environment. Pseudomonas aeruginosa is widely known to demonstrate ability in reducing copper concentration by using biofilm. In this study P. aeruginosa was employed to reduce copper concentration by culturing the bacteria in broth Luria-Bertani medium, supplemented with L-arginine at 80 mM concentration and CuSO4 at 750 ppm. The study demonstrated that the supplementation of medium using L-arginine increased biofilm formation by 52.41%, compared to to the treatment without L-arginine supplementation. Copper analysis also showed that P. aeruginosa was capable of reducing copper concentration by 145.54 ppm (20%). When the cell biomass was applied to biosorp copper in the CuSO4 solution at 750 ppm (without any medium supplementation), it was found that copper concentration in the solution was reduced by 155.18 ppm (21%), and 164.83 (23%) following incubation for 14 hours and 48 hours, respectively.
