Bioaktivitas Ekstrak Tunikata (Didemnum sp.)  dari Kultur Biorock di Bali sebagai Anti-Vibriosis dan Anti-Biofilm

Penelitian ini bertujuan untuk mengevaluasi bioaktivitas dari senyawa metabolit sekunder ekstrak sampel tunikata Didemnum sp. dari kultur Biorock sebagai anti-vibrio dan anti-biofilm. Sampel diambil dari kultur Biorock di Pemuteran, Bali, Indonesia. Penelitian dilakukan dengan beberapa pengujian, yaitu uji bioautografi, MIC (Minimum Inhibitory Concentration), MBC (Minimal Bactericidal Concentration), dan BIF (Biofilm Inhibitory Formation) dengan modifikasi. Kandungan senyawa dideteksi dengan reagen p-anesaldehida dan vanillin H2SO4. Uji bioautografi dilakukan dengan menggunakan larutan resazurin 0,01 µg/µl terhadap bakteri uji V. alginolyticus dan V. parahaemolyticus. Bioautografi menunjukkan adanya aktivitas senyawa terpenoid yang berperan pada aktivitas anti-vibrio. Uji MIC dilakukan dengan menggunakan 96 well plate dengan konsentrasi awal ekstrak sebesar 10 µg/µl. Uji MBC dilakukan dengan mengulturkan kembali hasil dari sumuran MIC terpilih pada media TBCS (Thiosulfate Citrate Bile Salt) agar. Hasil uji menunjukkan bahwa ekstrak sampel Didemnum sp. memiliki aktivitas anti-vibrio dengan nilai MIC sebesar 2,5 µg/µl. Hasil uji MBC menunjukkan bahwa senyawa metabolit sekunder ekstrak Didemnum sp. memiliki aktivitas bakteriostatik. Uji BIF menunjukkan ekstrak sampel Didemnum sp. tidak menghambat pembentukan biofilm pada V. alginolyticus dan V. parahaemolyticus.

Bioactivity Potency of Tunicate Extract (Didemnum sp.) from Biorock Culture in Bali as Anti-Vibriosis and Anti-Biofilm

 

                                                                                                                        Abstract

This study aims to evaluate the bioactivity of secondary metabolite compounds of crude extract of ascidian Didemnum sp.from Biorock culture as anti-vibrio and anti-biofilm. The sample was taken from Biorock culture in Pemuteran, Bali, Indonesia. The research was conducted with several tests, namely bioautography, MIC (Minimum Inhibitory Concentration), MBC (Minimum Bactericidal Concentration), and BIF (Biofilm Inhibitory Formation) tests with modifications. The compound content was detected with p-anesaldehyde and vanillin H2SO4 reagents. Bioautography test was conducted using 0.01 µg/µl resazurin solution against V. alginolyticus and V. parahaemolyticus. Bioautography showed the activity of terpenoid compounds that play a role in anti-vibrio activity. MIC test was conducted using 96 well plate with initial concentration of 10 µg/µl extract. MBC test was conducted by re-culturing the results of the selected MIC wells on TBCS (Thiosulfate Citrate Bile Salt) agar media. The test results showed that the Didemnum sp. sample extract had anti-vibrio activity with an MIC value of 2.5 µg/µl. MBC test results show that secondary metabolite compounds of Didemnum sp. extract has bacteriostatic activity. BIF test showed that Didemnum sp. sample extract did not inhibit biofilm formation on V. alginolyticus and V. parahaemolyticus.
