Isolation of Triterpenoids from Aglaia Foveolata as Antibacterial Agent Against Multidrug-Resistant Bacteria

antiinfeksi dilakukan. Kulit kayu Aglaia foveolata diisolasi untuk
mendapatkan senyawa murni. Antibakteri dengan Multidrug Resistance diuji
terhadap ekstrak tersebut untuk menemukan puncak-puncak aktivitas baru dengan
menggunakan MBC dan MIC. Dari hasil skrining, ekstrak menunjukkan fraksi mana
yang memiliki aktivitas antibakteri terhadap ekstrak murni. Senyawa tersebut
dianalisis menggunakan LC-HRMS, 1D NMR, FTIR, dan Spektrofotometri UV-Vis lalu
dibandingkan dengan database publik yang ada saat ini untuk senyawa yang sudah
ada.
Penelitian ini mengisolasi 17,24-Epoksi-25-hidroksibaccharan-3-on dari fraksi F8 sebanyak 345 mg berupa kristal putih, dan asam eiklerianat dari fraksi F15-SF10-F1 sebanyak 6.9 mg berupa padatan kuning. 17,24-Epoksi-25-hidroksibaccharan-3-on menunjukan aktivitas antibakteri terhadap Bascillus subtilis MDR dengan nilai MIC dan MBC sebesar 125 µg mL–1. Asam eiklerianat juga menunjukan aktivitas antibakteri pada Escherichia coli MDR dengan nilai MIC dan MBC sebanyak 125 µg mL–1.
Indonesia is a tropical country with high
biodiversity. Thus, there is a wide variety of plants of biological origin that
can be used as traditional medicinal ingredients. Antibiotics have been used
clinically to treat bacterial diseases since the 1940s. Unfortunately,
antimicrobial resistance is prevalent. The genus Aglaia is known for its
rich association of different species, often with interesting biological
activities. Tetracyclic triterpenes are known to be interesting bioactivities
in the genus Aglaia.
As a result, antiinfection screening was
performed. Aglaia foveolata bark was isolated in order to get a pure
compound. Antibacterial with MDR are tested against the extract to find new
peaks of action by using the MBC and MIC. From the screening, the extract
showed which fractions has antibacterial activity. The pure compound was
analyzed using LC-HRMS, 1D NMR, FTIR, and UV-Vis Spectrophotometry then
compared to the current public database for already existing compounds.
This research has
isolated 17,24-Epoxy-25-hydroxybaccharan-3-one from
fraction of F8 as much as 345 mg with a form of white in crystals, and
echlerianic acid from fraction of F15-SF10-F1 as much as 6.9 mg with a form of
yellow in solids. 17,24-Epoxy-25-hydroxybaccharan-3-one showed antibacterial
activity against Bacillus subtilis MDR with a MIC and MBC value of 125 µg mL–1. Echlerianic acid also showed antibacterial activity as well
in Escherichia coli MDR with a MIC and MBC value of 125 µg mL–1.
