Calcium hydroxide is the most popular root canal medicament used in endodontic treatment. A good root canal medicament is expected to inhibit biofilm formation in addition to its ability to inhibit bacterial growth. Garlic is effective in eliminating both gram-positive and gram-negative bacteria, so it can be used as an alternative root canal medicament. This study aimed to analyze the differences in the effect of garlic ethanol extract in various concentrations and calcium hydroxide on the formation of Pseudomonas aeruginosa ATCC 10145 biofilm.
Pseudomonas aeruginosa ATCC 10145 biofilm inhibition test by calcium hydroxide+glycerin, Calcigel® and garlic ethanol extract was carried out with a 96-well U bottom flexible Microplate. Garlic ethanol extract consists of several concentrations, namely concentrations of 80%, 40%, 20%, 10%, 5% and 2.5%. The test was carried out with five repetitions. The data obtained from inhibition of biofilms is in the form of optical density values. The optical density value is then used to calculate the percentage of inhibition. The percentage resistance results for each sample were analyzed using the Kruskal-Wallis statistical test, then followed by the U-Mann Whitney statistical test with a confidence level of 95%.
The test results showed that all ingredients were able to inhibit the formation of P.aeruginosa ATCC 10145 bacterial biofilm. Calcium hydroxide + glycerin had the largest inhibition percentage with an average inhibition percentage value of 101.12%. Garlic ethanol extract 80% has the greatest percentage of inhibition compared to other concentrations, namely 94.16%.
This research concludes that each treatment group can inhibit the formation of P. aeruginosa ATCC 10145 biofilm. The inhibitory activity of P. aeruginosa ATCC 10145 biofilm formation by garlic ethanol extract will increase with increasing concentration.
