ISOLASI DAN IDENTIFIKASI FRAKSI POLAR ETIL ASETAT DARI KULIT BATANG MAHONI (Aglaia foveolata) SEBAGAI ANTIBAKTERI

Isolation and identification of the polar ethyl
acetate fraction of mahogany (Aglaia foveolata) bark from the Mekongga
Forest, Southeast Sulawesi, has been carried out. This study aims to determine
the antibacterial potential of the crude extract, fraction, and pure isolate of
the Aglaia foveolata plant against Gram positive and negative bacteria. This research was initiated by extracting the stem
bark of Aglaia foveolata and then successively partitioning it using
solvents of different polarity, namely n-hexane, ethyl acetate, and butanol.
These fractions were tested as antibacterial to find out which fractions gave the
best activity. The selected fractions were separated by chromatographic
methods, namely thin layer chromatography, gravity column chromatography, and
preparative high performance liquid chromatography. The pure isolates obtained
were characterized to determine their chemical structure by various
spectroscopic methods including UV-Vis, FTIR, high performance liquid
chromatography tandem high resolution mass spectroscopy (LC-HRMS), 1D NMR, and
2D NMR. Antibacterial testing of pure isolates was carried out on MDR
(Multidrug-resistance) strains of bacteria using the broth microdilution assay
method. Two isolates obtained as pure compounds based on
UV-Vis, FTIR, LC-HRMS, and NMR analysis were identified as
17,24-epoxy-25-hydroxy-3-oxobaccharan-21-oic acid (1) and
stigmasterol-5,22 -diene-3-O-?-D-glucopyranoside (2). Isolate 1 showed
bioactivity against Gram negative bacteria (E. coli) with MIC 62.5 µg/mL
and Gram positive (S. aureus) 125 µg/mL. Isolate 2 showed bioactivity
against Gram positive bacteria (S.aureus and B. subtilis) with an
MIC of 250 µg/mL and gram negative bacteria (E. coli, P. aeruginosa,
and
K. pneumonia) with an MIC value of 250 µg/mL.
