Lactic acid
bacteria (LAB) can be isolated from fermented Ambon banana peel liquid. LAB
have bacteria potential to produce metabolites to inhibit the growth of the
pathogenic bacterium Xanthomonas campestris (X. campestris) in cabbage, tomato, and strawberry
plants. LAB is known to inhibit the growth of X. campestris which causes
black spots on plants which results in a decrease in productivity. This study is
aims to obtain LAB isolates from fermented Ambon banana peel liquid, select LAB
isolates that have the potential as X. campestris antibacterials, and
characterize LAB isolates based on their biochemical properties. From the
fermented banana peel liquid that added molasses with a
ratio of 9:1, sixteen isolates were obtained which were
then characterized to ensure that the isolates obtained belonged to the LAB
group using gram staining, morphological, catalase, motility, and cellulose
degradation tests. After that, an antibacterial test was carried out using the
well method and followed by biochemical tests including the indole, carbohydrate
fermentation, and gas production tests to identify the LAB genus. From the X.
campestris antibacterial test, four isolates were selected which produced
clear zones with an average diameter of 3.72-5.76 mm and belonged to the weak
to moderate category. The isolates of then tested for biochemistry analysis and
the results showed positive fermentation of glucose, sucrose, and lactose. In
the gas production test, the results showed that the isolate belonged to the
homofermentative type. The indole test, have a negative result was obtained
where no indole ring was formed. Based on the results of this study, four LAB
isolates isolated from Ambon banana peel fermented liquid including to gram
positive, catalase negative, negative motility, negative cellulose degradation and were
known to have the ability to inhibit the growth of X. campestris which
could then be used as raw material for biofertilizers.
