Uji Aktivitas Antibakteri dan DPPH Radical Scavenging Ekstrak Etanol Benalu Teh (Scurrula atropurpurea) secara In Vitro terhadap Cutibacterium acnes sebagai Agen Antijerawat dalam Serum

Benalu teh (Scurrula atropurpurea) is known to have inhibitory activity against several types of bacteria. However, no studies have shown its activity against Cutibacterium acnes bacteria. This bacterium plays a role in the pathophysiology of acne vulgaris (AV). This study aims to determine the antioxidant and antibacterial activity of Ethanol Benalu Tea Extract (EEBT) as a stable anti-acne agent in a liquid topical formula.
EEBT was obtained by macerating benalu teh with 70% ethanol. The macerate was then freeze dried to obtain EEBT in powder form. This study analyzed the total phenolic content by Folin-Ciocalteu, DPPH radical scavenging activity by microdilution, and antibacterial activity of EEBT. Antibacterial activity test was conducted using microdilution method with BHI media against C. acnes bacteria. EEBT was then formulated into a topical solution preparation, hereafter referred to as serum, and analyzed for pH stability and viscosity using the 5-cycle freeze-thaw method.
EEBT which produced an extraction yield of 12.32% had a total phenolic content of 11.129 ± 0.190 mgGAE/g. From this study, it is known that EEBT has very strong antioxidant activity with IC50 DPPH radical scavenging of 24,20 ppm. EEBT also has potential as an anti-acne agent with MIC50 of 5.731 mg/mL against C. acnes. EEBT serum is known to have decreased viscosity after 5 cycles of freeze-thaw testing but still within the viscosity acceptance range. Serum EEBT experienced a decrease in pH to 3.91 which is still close to the acceptance value of topical preparations, namely 4.5-6.5. Formula optimization is needed, especially optimization of the concentration of HPMC, propylene glycol, tween 80, and PEG400 used in order to obtain a more stable serum and good physical characteristics. In addition, antibacterial activity testing of the serum is needed to ensure the success of AV therapy with EEBT serum.
