POTENTIAL USE OF BOVINE UMBILICAL VEIN ENDOTHELIAL CELLS CONDITIONED MEDIUM (BUVECs-CM) IN HEALING BURNS: AN OVERVIEW OF BLOOD VESSELS

Bovine Umbilical Vein Endothelial Cells (BUVEC)adalah sel primer yang diisolasi langsung dari vena umbilikalis sapi.BUVEC adalah sel yang melekat yang memiliki morfologi sel seperti epitel, yang berbentuk poligonal dengan nukleus oval hingga bulat di tengah sel. embuluh darah berperan penting dalam penyembuhan luka terutama melindungi luka. Penelitian ini menggunakan 48 ekor tikus Wistar (Sprague Dawley) jantan berumur 2-3 bulan dengan berat badan 180-220 gram. Bahan yang digunakan untuk tikus uji adalah Bovine Umbilical Vein Endothelial Cells-Conditioned Medium (BUVEC-CM) berupa BUVEC- CM 5%, 10%, 15% krim BUVEC-CM. Metode penelitian yang digunakan adalah persiapan perawatan luka, persiapan model hewan, pembuatan luka, eutanasia, persiapan jaringan pengambilan sampel dan pewarnaan Hematoxylin Eosin (HE). Dalam percobaan yang kami lakukan ini, krim BUVEC-CM digunakan sebagai obat untuk menyembuhkan luka. Krim dioleskan dua kali sehari dari hari 1 sampai 29. Hasil yang dikumpulkan adalah dari hari 5, 13, 21 dan 29. Dua belas ekor tikus dari masing-masing kelompok dikorbankan pada hari ke 5, 13, 21 dan 29 setelah dilukai. Penutupan luka telah digunakan untuk memantau penyembuhan luka. Kesimpulannya, krim BUVEC-CM merupakan krim efektif yang dapat digunakan untuk menyembuhkan luka bakar dan pertumbuhan pembuluh darah yang semakin meningkat.

Bovine Umbilical Vein Endothelial Cells (BUVEC) are primary cells isolated directly from the umbilical vein of cattle. BUVEC is an adherent cell that has an epithelial-like cell morphology, which is polygonal in shape with an oval to round nucleus in the center of the cell.Blood vessels play an important role in wound healing  especially, protecting the wound. This study uses 48 male Wistar rats (Sprague Dawley) ranging from 2-3 months and weighs 180-220 grams.Materials used for the tested rats are Bovine Umbilical Vein Endothelial Cells-Conditioned Medium (BUVEC-CM) in the form of BUVEC-CM 5%, 10%, 15 % of BUVEC-CM cream. The method of research are preparation of wound treatment, preparation of animal model, wound creation, euthanasia, sample collection tissue preparation and Hematoxylin Eosin (HE) staining.In this experiment that we conducted, the BUVEC-CM cream was used as a medicine to heal the wounds. The cream was applied twice everyday from day 1 to 29. the results collected were from day 5, 13, 21 and 29. Twelve rats from each group were sacrificed at day 5, 13, 21 and 29 after wounded. Wound closure has been used to monitor wound healing. In conclusion, the BUVEC-CM cream is an effective cream that can be used to heal burn wounds and the growth of blood vessels increases progressively.
