Potential Use of Bovine Umbilical Vein Endothelial Cells Conditioned Medium (BUVEC-CM) in Wound Healing: An Overview of the Role of Collagen

Bovine Umbilical Vein Endothelial Cells (BUVEC-CM) merupakan sel primer yang diisolasi langsung dari pembuluh darah vena umbilikus sapi. BUVEC merupakan adherent sel yang memiliki morfologi epithelial-like cell, yaitu berbentuk poligonal dengan inti oval hingga bulat yang berada di tengah sel. Conditioned Medium (CM) merupakan medium yang dikoleksi dari kultur primer sel tertentu setelah dikultur selama beberapa hari. Luka bakar adalah cedera (injury) sebagai akibat kontak langsung atau terpapar dengan sumber-sumber panas, listrik, zat kimia, atau radiasi. Penelitian ini bertujuan untuk menganalisis potensi aplikasi krim Bovine Umbilical Vein Endothelial Cells-Conditioned Medium (BUVEC-CM) pada proses kesembuhan luka bakar tipe II dengan parameter kualitatif penutupan luka dari segi pembentukan kolagen dengan mengevaluasi sampel kulit tikus secara histologis dengan pewarnaan Masson’s Trichrome Staining (MTS). 48 tikus Wistar jantan dibagi 4 kelompok, dengan masing-masing kelompok ada 12 ekor tikus. Luka bakar tipe II dilakukan pada setiap tikus dan dirawat dengan povidone iodine dan krim BUVEC-CM konsentrasi 5%,10?n 15%. Pada hari ke 5, 13, 21, 29 tikus dieutanasia dan diambil sampel kulit yang di fiksatif dalam formalin. Kesembuhan luka bakar dari segi konformasi kolagen yang di ekstensi ke lapisan dermis dianalisis dengan pewarnaan MTS untuk mengetahui potensi krim BUVEC-CM 5, 10, 15% terhadap penutupan luka dalam interval lapan hari.

Bovine Umbilical Vein Endothelial Cells (BUVEC-CM) are primary cells isolated directly from the umbilical veins of cattle. BUVEC is an adherent cell that has an epithelial-like cell morphology, which is polygonal in shape with an oval to round nucleus in the center of the cell. Conditioned Medium (CM) is a medium that is collected from the primary culture of certain cells after being cultured for several days. Burns are injuries that results from direct contact or exposure to sources of heat, electricity, chemicals, or radiation. This study aims to analyze the potential application of Bovine Umbilical Vein Endothelial Cells-Conditioned Medium (BUVEC-CM) cream in the healing process of Type II burns with qualitative parameters of wound closure in terms of collagen formation.by evaluating skin samples with Masson’s Trichrome Staining (MST). 48 male Wistar rats were divided into 4 groups, with 12 rats in each group. Type II burns were performed on each rat and treated with povidone iodine and BUVEC-CM cream with concentrations of 5%, 10% and 15%. On day 5, 13, 21, 29 rats were euthanized and skin samples were taken which were fixed in formalin. Collagen formation in the healing of burns extended to the dermis layer was analyzed by MTS to determine the potency of BUVEC-CM 5, 10, 15% cream for wound closure in the interval of 8 days.
