Microbial contamination is a problem often faced by traditional medicine industries. Medicinal plants are highly vulnerable to microbial contamination, thus requiring appropriate decontamination methods. Ozone is an excellent oxidizing agent that can be utilized to kill bacteria, making this method potentially suitable for use on medicinal plant materials. The aim of this research is to determine the effect of microbial decontamination using ozone on the rhizome of fingerroot (Boesenbergia rotunda (L.) Mansfeld).
Samples were treated with ozone for 15, 30, 60, and 120 minutes with an ozone generator machine with a production capacity of 10 g/hour (10G).. The contamination levels of microorganisms were examined using the total plate count (TPC) and mold and yeast count (MYC) tests, the pinostrobin content was determined using thin-layer chromatography (TLC)-densitometry, and the free radical scavenging activity was assessed using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging test.
The results of the analysis obtained from this test revealed that the optimal ozone dosage to reduce microbial contamination levels in the simplicia of temu kunci is ozone treatment for 120 minutes, resulting in a decrease of 92.67% in TPC value, 84.17% in MYC value, 1.27% in pinostrobin content, and relatively unchanged activity in free radical scavenging.
