POTENSI RIMPANG TEMU GIRING (Curcuma heyneana Val. & Zijp) SEBAGAI AGEN KEMOPREVENTIF PADA KANKER HATI MENGGUNAKAN MODEL SEL JHH-4

Hepatocellular carcinoma cells are the most common type of liver cancer. Curcuminoids are natural polyphenol compounds that are abundant in Curcuma heyneana ethanolic Extract (ETG) and are known to inhibit cancer cell proliferation. This study aims to reveal the potential of Curcuma heyneana Extract (ETG) as a chemopreventive agent in liver cancer using the JHH-4 cell as a model.
ETG was obtained by maceration method using ethanol which was then identified for its phytochemical profile using Thin Layer Chromatography (TLC). Then the TLC results were quantified to calculate the levels of compounds present in the ETG based on spot intensity with the ImageJ application. The DPPH Antioxidant Test was conducted to determine the antioxidant activity of ETG in the radical scavenging mechanism. The cytotoxic activity of ETG on JHH-4 liver cancer cells was tested by the MTT Assay.
Extraction produces a yield of 10,2 %w/w. ETG contains a curcuminoid compound of 4.52 %w/w consisting of curcumin, demethoxycurcumin, bisdemethoxycurcumin with successive levels of 0.49 %w/w; 3.21 %w/w; and 0.82%w/w. ETG had antioxidant activity in the DPPH radical scavenger test with an IC50 of 378.96 ?g/mL with ascorbic acid (Vitamin C) as a positive control having an IC50 of 8.49 ?g/mL. The single cytotoxic activity of ETG on JHH-4 cells has an IC50 value of 16.62 ?g/mL which is classified as having strong cytotoxic activity. This study concluded that ETG has the potential to be developed as a chemopreventive agent in liver cancer.
